The Biological Effects of Dickkopf1 on Small Cell Lung Cancer Cells and Bone Metastasis

Hailin Pang1, Ningqiang Ma1, Mi Jiao, Weiwei Shen, Bo Xin, Tongfei Wang, Feng Zhang, Lili Liu, Helong Zhang

    Oncology Research, Vol.25, No.1, pp. 35-42, 2017, DOI:10.3727/096504016X14719078133249

Abstract The bone is among the most common sites of metastasis in patients with lung cancer. Over 30%–40% of lung cancers can develop bone metastasis, and no effective therapeutic methods exist in clinic cases. Wnt/β-catenin signaling and Dickkopf1 (DKK1) play important roles in the progression of lung cancer, which preferentially metastasizes to the skeleton. However, the role of DKK1 in osteotropism of small cell lung cancer (SCLC) remains to be elucidated. This study aimed to define the role of DKK1 in SCLC bone metastasis and investigate the underlying mechanisms. Our results demonstrated that the expression level… More >

MicroRNA-200a Suppresses Cell Invasion and Migration by Directly Targeting GAB1 in Hepatocellular Carcinoma

Jianlin Wang*1, Wenjie Song*1, Weiwei Shen†1, Xisheng Yang*, Wei Sun*, Sshibin Qu*, Runze Shang*, Ben Ma*, Meng Pu*, Kaishan Tao*, Kefeng Dou*, Haimin Li*

    Oncology Research, Vol.25, No.1, pp. 1-10, 2017, DOI:10.3727/096504016X14685034103798

Abstract MicroRNA-200a (miR-200a) is frequently downregulated in most cancer types and plays an important role in carcinogenesis and cancer progression. In this study, we determined that miR-200a was downregulated in hepatocellular carcinoma (HCC) tissues and cell lines, consistent with the results of our previous study. Because a previous study suggested that downregulation of miR-200a is correlated with HCC metastasis, we aimed to elucidate the mechanism underlying the role of miR-200a in metastasis in HCC. Here we observed that overexpression of miR-200a resulted in suppression of HCC metastatic ability, including HCC cell migration, invasion, and metastasis, in More >

    Canadian Journal of Urology, Vol.23, No.2, pp. 8227-8233, 2016

Abstract Introduction: To compare surgical complications and tyrosine kinase inhibitor (TKI)-toxicities in patients who underwent primary cytoreductive nephrectomy (CN) followed by adjuvant TKI therapy versus those who underwent neoadjuvant TKI therapy prior to planned CN for metastatic renal cell carcinoma (mRCC).
Materials and methods: Two-center retrospective analysis. Sixty-one mRCC patients underwent TKI therapy with sunitinib between July 2007 to January 2014. Patients were divided into three groups: primary CN followed by adjuvant TKI (n = 27, Group 1), neoadjuvant TKI prior to CN (n = 21, Group 2), and primary TKI alone (no surgery, n = 13, Group… More >

Bladder cancer will grow anywhere: report of a urothelial carcinoma drop metastasis to the vagina and literature review

    Matthew A. Uhlman1, Mark D. Bevill1, Michael J. Goodheart2, James A. Brown1, Michael A. O’Donnell1

    Canadian Journal of Urology, Vol.23, No.4, pp. 8379-8381, 2016

Abstract Urothelial carcinoma is the 2nd most common cancer of the urinary tract and accounts for the majority of cases of bladder cancer. Metastases are not infrequently encountered, increasing with disease stage and are most commonly seen in the bones and lungs. Many other sites have been described including the omentum, liver, and ovaries. An extremely rare site of metastatic disease however is within the vagina. Here we present a case of a probable vaginal “drop metastasis” from previously treated urothelial carcinoma in the ureter and bladder. More >

Inhibition of Proliferation, Migration, and Invasion by Knockdown of Pyruvate Kinase-M2 (PKM2) in Ovarian Cancer SKOV3 and OVCAR3 Cells

Yi Miao1, Meng Lu1, Qin Yan, Shuangdi Li, Youji Feng

    Oncology Research, Vol.24, No.6, pp. 463-475, 2016, DOI:10.3727/096504016X14685034103671

Abstract Pyruvate kinase (PK) is a key enzyme in the process of glycolysis, catalyzing phosphoenolpyruvate (PEP) into pyruvate. Currently, PK isozyme type M2 (PKM2), one subtype of PK, has been proposed as a new tumor marker with high expression in various tumor tissues. Here we aimed to explore the effects of siRNAPKM2 on ovarian carcinoma (OC) cell lines SKOV3 and OVCAR3, in which PKM2 was notably expressed. PKM2 gene interference lentivirus vectors were built by miRNA transfection assay. siRNA-PKM2-transfected SKOV3 and OVCAR3 cells were evaluated for cell proliferation, cell cycle distribution, cell apoptosis, cell migration, and More >

Knockdown of Long Noncoding RNA uc.338 by siRNA Inhibits Cellular Migration and Invasion in Human Lung Cancer Cells

Xuexin Gao*, Xuezhen Gao, Chao Li*, Yukun Zhang*, Lei Gao

    Oncology Research, Vol.24, No.5, pp. 337-343, 2016, DOI:10.3727/096504016X14666990347671

Abstract Lung cancer remains a critical health concern worldwide. Long noncoding RNAs with ultraconserved elements have recently been implicated in human tumorigenesis. The present study investigated the role of ultraconserved element 338 (uc.338) in the regulation of cell proliferation and metastasis in human lung cancer. Our data showed that the expression of uc.338 in lung cancer was remarkably increased in vivo and in vitro. Depletion of uc.338 with specific siRNA interference retarded the cell proliferative rate in lung cancer cell lines NCI-H929 and H1688. Furthermore, knockdown of uc.338 caused cell cycle arrest in the G0/G1 phase in More >

Knockdown of Long Noncoding RNA PCAT6 Inhibits Proliferation and Invasion in Lung Cancer Cells

    Li Wan1, Lin Zhang1, Kai Fan, Zai-Xing Cheng, Quan-Chao Sun, Jian-Jun Wang

    Oncology Research, Vol.24, No.3, pp. 161-170, 2016, DOI:10.3727/096504016X14618564639178

Abstract As a newly identified oncogenic long noncoding RNA (lncRNA), prostate cancer-associated transcript 6 (PCAT6) promoted cellular proliferation and colony formation of prostate cancer. However, the biological function of PCAT6 in lung cancer is still largely unknown. In this study, we found that PCAT6 is significantly increased in cancer tissues compared to normal tissues and positively correlates with metastasis of lung cancer in patients. We then examined PCAT6 expression in lung cancer cell lines and identified that PCAT6 expression was significantly elevated in lung cancer cells compared to normal human bronchial epithelial (NHBE) cells, especially in… More >

    Scott M. Norberg1, Michelle Oros1, Varsha Manucha2, Daniel Eun1, Marijo Bilusic3

    Canadian Journal of Urology, Vol.22, No.2, pp. 7755-7757, 2015

Abstract Cutaneous metastases from urologic cancers are very uncommon, usually represent widespread metastatic disease and are associated with a very poor prognosis. They may occur in 1% of patients with urologic malignancies, most commonly from kidney, followed by bladder and prostate tumors. In this report, we describe a case of urothelial carcinoma with metastases to the scrotum treated with platinum based chemotherapy with a durable complete response lasting more than 14 months. Molecular profiling revealed deleterious mutations in e-cadherin and retinoblastoma genes, suggesting their possible role in the pathogenesis of cutaneous metastases. Further studies are needed More >

Symptomatic cardiac metastasis responding to pazopanib in a patient with renal cell carcinoma

Ramin Behzadigohar, Mark A. Rosenthal, Ben Tran

    Canadian Journal of Urology, Vol.22, No.3, pp. 7824-7826, 2015

Abstract We report a case of an 82-year-old man with renal cell carcinoma who developed a cardiac metastasis within the interventricular septum. He had been under watchful waiting for indolent metastatic renal cell carcinoma for many years before developing symptoms consistent with heart failure. At this time, a 44 mm interventricular septal mass, consistent with a cardiac metastasis, was identified as the cause of his symptoms. Pazopanib was initiated which led to both a clinical and radiological response. More >
