Abstract Introduction: Chemokine-like factor 1 (CKLF1) is a chemokine that is overexpressed in several diseases. Our previous findings revealed a significant increase in CKLF1 expression in the ischemic brain, suggesting its potential as a therapeutic target for ischemic stroke. Methods: In this study, we examined the expression dynamics of CKLF1 in both in vivo and in vitro models of ischemic cardiac injury. Myocardial infarction (MI) was induced in vivo by ligation of the left anterior descending artery (LAD) of the rat heart. The levels of CKLF1, Creatine Kinase MB Isoenzyme (CK-MB), and Lactate dehydrogenase (LDH) in the serum were… More > Graphic Abstract

OPA3 overexpression modulates lipid droplet production and sensitizes colorectal cancer cells to bevacizumab treatment

    HONGBIAO WU*, DONGFANG LIU

    BIOCELL, Vol.48, No.6, pp. 971-980, 2024, DOI:10.32604/biocell.2024.049466 - 10 June 2024

Abstract Background: Colorectal cancer (CRC) represents a substantial risk to public health. Bevacizumab, the first US FDA-approved antiangiogenic drug (AAD) for human CRC treatment, faces resistance in patients. The role of lipid metabolism, particularly through OPA3-regulated lipid droplet production, in overcoming this resistance is under investigation. Methods: The protein expression pattern of OPA3 in CRC primary/normal tissues was evaluated by bioinformatics analysis. OPA3-overexpressed SW-480 and HCT-116 cell lines were established, and bevacizumab resistance and OPA3 effects on cell malignancy were examined. OPA3 protein/mRNA expression and lipid droplet-related genes were measured with Western blot and qRT-PCR. OPA3… More > Graphic Abstract

    BIOCELL, Vol.48, No.2, pp. 205-216, 2024, DOI:10.32604/biocell.2023.045109 - 23 February 2024

Abstract Introduction: Dexamethasone (Dex) caused impaired osteoblast differentiation and oxidative stress (OS) in bone marrow mesenchymal stem cells (BMSCs). This work sought to elucidate the precise molecular pathway through which Dex influences osteogenic differentiation (OD) and OS in BMSCs. Methods: The expression of Runt-related transcription factor 1 (RUNX1) and alpha-2 macroglobulin (A2M) was assessed in Dex-treated BMSCs using qRT-PCR and Western Blot. Following the functional rescue experiments, cell proliferation was determined by MTT assay, reactive oxygen species (ROS) expression by DCFH-DA fluorescent probe, lactate dehydrogenase (LDH), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (Gpx) expression… More >

Abstract Castor is one of the top 10 oil crops in the world and has extremely valuable uses. Castor inflorescences directly affect yield, so the study of inflorescence development is very important in increasing castor yield. Our previous studies have shown that the PIP5K gene family (PIP5Ks) is associated with inflorescence development. In this study, to determine the function of each PIP5K gene in castor, a female Lm-type castor line, aLmAB2, was used to determine the relative expression levels of the PIP5Ks in castor inflorescences. Six PIP5K genes were heterologously overexpressed in Arabidopsis thaliana, the relative expression of each gene… More >

    Phyton-International Journal of Experimental Botany, Vol.92, No.11, pp. 3063-3075, 2023, DOI:10.32604/phyton.2023.043803 - 24 October 2023

Abstract Adventitious root formation poses a major constraint on the tissue culture and genetic transformation of Eucommia ulmoides. Micrografting offers a new method for the transplantation of genetic transformation, and its success depends on the formation of graft unions. This study used transgenic rootless test-tube seedlings as scions and seedlings from seed as rootstocks during micrografting to avoid the rooting issues that occur during tissue culture and to investigate the role of the EuEG1 gene in the graft healing process. We found that the EuEG1 gene is a vital regulator of graft, and its overexpression contributes to the… More >

Ectopic Overexpression of EuCHIT30.7 Improves Nicotiana tabacum Resistance to Powdery Mildew

    Lanjun Li1, Degang Zhao1,2,*

    Phyton-International Journal of Experimental Botany, Vol.92, No.11, pp. 3043-3061, 2023, DOI:10.32604/phyton.2023.031175 - 24 October 2023

Abstract Various strains of powdery mildew (PM), a notorious plant fungal disease, are prevalent and pose a significant threat to plant health. To control PM, transgenic technology can be used to cultivate more resistant plant varieties. In the present study, we utilized the rapid amplification of cDNA ends (RACE) technique to clone the full-length cDNA sequence of the EuCHIT30.7 gene to explore plant genes with disease resistance functions. Bioinformatics analysis revealed that this gene belongs to the GH18 family and is classified as a class III chitinase. The EuCHIT30.7 gene is expressed throughout the Eucommia ulmoides plant, with… More >

Abstract Drought has severely affected the yield and quality of commercial crops. The BRI1 family plays an important role in plant response to drought stress, and BRL3 gene plays an important role in the study of drought in Arabidopsis thaliana. In this study, NtBRL3 was constructed as a vector and genetically transformed to obtain ‘N. Tobacco K326’ overexpression of NtBRL3. The enzyme activities of transgenic tobacco and wild-type tobacco were measured and transcriptome and metabolome analyses were performed. The results showed that the antioxidant enzymes of transgenic tobacco were more active under drought conditions, and 85 significantly differentially metabolites and More >

Abstract The auxin polar transporter, PIN-FORMED 2 (PIN2) plays an important role in root development. However, it remains unclear whether PIN2 genes form two Liriodendron species, L. chinense (LcPIN2) and L. tulipifera (LtPIN2), are both involved in root development and whether and to what extent these two genes diverge in function. Here, we cloned and overexpressed LcPIN2 and LtPIN2 in Arabidopsis thaliana wild-type (WT) and Atpin2 mutant. Phylogenetic and sequence analysis showed a small degree of differentiation between these two Liriodendron PIN2 genes. Tissue-specific gene expression analysis indicated that both Liriodendron PIN2 genes were highly expressed in roots, implying a potential role in root… More >

Abstract Chrysanthemum morifolium Ramat is one of the four major cut flowers in the world. Pht1 family is focus on the uptake and transport of phosphate in plants. In our previous studies, CmPht1;2 overexpression line (Oe1) had higher phosphate contents both in roots and shoots, and its root development was significantly enhanced than wild type (WT) at low phosphorus conditions in chrysanthemum. Metabolomics analysis showed that several metabolites had a change in pyruvate metabolism and tricarboxylic acid (TCA) cycle pathway. To explore the gene difference expression and the change of metabolic pathway between CmPht1;2-Oe1 and WT, we conducted… More >

Abstract Calmodulin (CaM) proteins play a key role in signal transduction under various stresses. In the present study, the effects of a sugarcane ScCaM gene (NCBI accession number: GQ246454) on drought and salt stress tolerance in transgenic Arabidopsis thaliana and Escherichia coli cells were evaluated. The results demonstrated a significant negative role of ScCaM in the drought and salt stress tolerance of transgenic lines of A. thaliana, as indicated by the phenotypes. In addition, the expression of AtP5CS and AtRD29A, two genes tightly related to stress resistance, was significantly lower in the overexpression lines than in the wild type. The growth More >
