Abstract Gastric cancer is the fourth most common type of cancer and the second highest leading cause of cancer-related deaths worldwide. It has already been established that miR-133a is involved in gastric cancer. In this study, we investigated the molecular mechanisms by which miR-133a inhibits the proliferation of gastric cancer cells. We analyzed the proliferative capacity of human gastric cancer cells SNU-1 using an MTT assay. Cell apoptosis was determined using flow cytometry. The expression levels of ERBB2, p-ERK1/2, and p-AKT in SNU-1 cells were determined using Western blot analysis. To confirm that ERBB2 is a direct target of miR-133a, a… More >

The expression and function of miR-376a-3p/DLX axis in gastric cancer cells

YAN ZHANG, CHAOJUN ZHANG*, ZHEN CAO, ZHANWEI ZHAO, LIN CHEN*

    BIOCELL, Vol.46, No.9, pp. 2073-2080, 2022, DOI:10.32604/biocell.2022.020635

Abstract Gastric cancer (GC) was referred to a malignant tumor of the digestive tract originating from the epithelium of gastric mucosa. Transcription factor DLX5 was verified as an oncogene in various types of tumors, while miR-376a-3p was speculated as a tumor suppressor. Based on the bioinformatics database, we hypothesized that miR-376a participated in the regulation of GC development by targeting DLX5. Compared with adjacent tissue, a significant increase of DLX5 expression was determined in GC tissues, but the expression level is significantly reduced in miR-376a. Similar expression signature of DLX5 and miR-376a was also determined between 4 GC cells (HGC, SGC,… More >

Arsenic trioxide inhibits the activity of SphK1 by decreasing the level of phosphatidylserine and phosphatidic acid in the human gastric cancer cell line MGC-803

PEI LIU, ZHE CHEN, XIAOQIONG MA*

    BIOCELL, Vol.46, No.3, pp. 737-743, 2022, DOI:10.32604/biocell.2021.015786

Abstract Sphingosine kinase 1 (SphK1) is an important synthetase during the synthesis of sphingosine-1-phosphate (S1P) from sphingosine (Sph). Previous studies demonstrated that arsenic trioxide (As2O3) could reduce the level of S1P in human gastric cancer cell line MGC-803, indicating that As2O3 may inhibit the activity of SphK1. In this study, the effect of As2O3 on the SphK1 activation pathway was investigated. Western blot and quantitative real-time PCR analysis were used to evaluate the changes in protein and mRNA levels. The multi-dimensional mass spectrometry-based shotgun lipidomics method (MDMS-SL) was used for the quantitative detection of phosphatidylserine (PS) and phosphatidic acid (PA). The… More >

    BIOCELL, Vol.46, No.3, pp. 721-735, 2022, DOI:10.32604/biocell.2021.015867

Abstract The peritoneum is the most common site of recurrence of gastric cancer (GC). Early occult peritoneal metastasis is difficult to detect by imaging examination. Stratifying the risk of peritoneal metastasis in patients with different Lauren subtypes is of great clinical value. We performed a univariate Cox regression to identify those genes with prognostic value of overall survival (OS) and peritoneal-specified disease-free survival (psDFS) from the Gene Expression Omnibus database. The candidate genes were screened by the Subpopulation Treatment Effect Pattern Plot (STEPP) method. Propensity score matching (PSM) analysis was used to reduce the interference of confounders on the results. Based… More >

Construction and validation of prognostic model based on autophagy-related lncRNAs in gastric cancer

    MENGQIU CHENG1,2, WEI CAO2, GUODONG CAO1, XIN XU1,2,*, BO CHEN1,*

    BIOCELL, Vol.46, No.1, pp. 97-109, 2022, DOI:10.32604/biocell.2021.015608

Abstract Gastric cancer (GC) is one of the most common cancer worldwide. Although emerging evidence indicates that autophagy-related long non-coding RNA (lncRNA) plays an important role in the progression of GC, the prognosis of GC based on autophagy is still deficient. The Cancer Genome of Atlas stomach adenocarcinoma (TCGA-STAD) dataset was downloaded and separated into a training set and a testing set randomly. Then, 24 autophagy-related lncRNAs were found strongly associated with the survival of the TCGA-STAD dataset. 11 lncRNAs were selected to build the risk score model through the least absolute shrinkage and selection operator (LASSO) regression. Every patient got… More >

The Functions of MicroRNAs and Their Potential Applications in the Diagnosis and Treatment of Gastric Cancer

    Yongxia He1, Zheng Wang1, Yue Wang2, Man Sun3,*

    Oncologie, Vol.23, No.3, pp. 351-357, 2021, DOI:10.32604/Oncologie.2021.014772

Abstract Gastric cancer is a highly malignant disease with complex pathogenic mechanisms, and has high incidence and mortality rate. At present, the diagnosis of gastric cancer mainly includes gastroscopy, serum analysis and needle biopsy, and the treatment methods include conventional surgical resection, radiotherapy and chemotherapy. Yet, some limitations were involved in these diagnostic and therapeutic methods, so accurate targeted therapy has received considerable attention. MicroRNAs (miRNAs) are non-coding RNA that can interact with the 3-terminal non-translational region of the target gene mRNA to reduce the expression of the target gene, participate in the regulation of multiple signaling pathways, and play an… More >

    BIOCELL, Vol.45, No.6, pp. 1521-1526, 2021, DOI:10.32604/biocell.2021.09220

Abstract Increasing evidence proves that circular RNAs (circRNAs) play an important role in regulating the biological behaviors of tumors. The central purpose of this research was to investigate the functions of circRNA in gastric cancer. The utilization of real-time PCR was to test circPTN expression in gastric cancer cells. Cell counting colony formation assays, CCK-8 assay, and EdU assay were used to investigate proliferation. Transwell assay was applied to investigate migration. We discovered that circPTN was highly expressed in gastric cancer cells. Low expression of circPTN inhibits gastric cancer cell proliferation and migration. Elevated expression of circPTN promotes gastric cancer cell… More >

Abstract Helicobacter pylori infection has been significantly linked to Peptic Ulcer Disease and Gastric Cancer. Metabolomic fingerprinting may offer a principal way of early diagnosis and to understand the molecular mechanism of H. pylori-induced pathogenicity. The rationale of the study is to explore the underlying distinct metabolic mechanisms of H. pylori-induced PUD and to identify potential biomarkers for disease diagnosis and associated risks using Gas chromatography/mass spectrometry. GC/MS-based analytical method was used to compare metabolic profiles of healthy controls (N = 20) and peptic ulcer patients (N = 45). Acquired metabolomic data were analyzed by constructing a diagnostic model using principal… More >

    BIOCELL, Vol.45, No.4, pp. 911-921, 2021, DOI:10.32604/biocell.2021.010119

Abstract Distal gastric cancer (DGC) is a subgroup of gastric cancer (GC), which has different molecular characteristics from proximal gastric cancer (PGC). These differences result in different overall survival (OS) rates; however, data pertaining to the survival rate in PGC or DGC are contradictory. This suggests that the location of GC is not the unique cause of the different survival rates, while the molecular characteristics might be more important factors determining the prognosis of DGC. Therefore, the aim of this study was to discover key prognostic factors in DGC using bioinformatic methods and to explore the potential molecular mechanism. The Cancer… More >
