    Phyton-International Journal of Experimental Botany, Vol.81, pp. 143-148, 2012, DOI:10.32604/phyton.2012.81.143

Abstract The objective of this research was to increase the genetic diversity of the Nopal Germplasm Bank of the FAUANL using RAPD molecular markers to estimate the genetic diversity of 15 accessions of cactus pear from molecular markers recorded in the Bank. DNA extraction was performed by the method of cellular ruptor using FastDNA® Kit. Performance measurement of DNA was carried out through Picoflour. It was observed that the DNA concentrations ranged from 74.91 ng/µL to 12.32 ng/uL. Twelve primers were used to generate from 1 to 5 bands per primer with a molecular weight ranging from More >

Obtaining new germplasm in Cenchrus ciliaris L. through induced-mutation and in vitro selection

    López Colomba E1, A Prina2, S Griffa1, AN Ribotta1, E Carloni1, E Tommasino1, C Luna1, E Biderbost1, K Grunberg1

    Phyton-International Journal of Experimental Botany, Vol.80, pp. 59-64, 2011, DOI:10.32604/phyton.2011.80.059

Abstract Cenchrus ciliaris L., a forage grass of wide distribution in the north-west of Argentina, is a tetraploid (4x = 36) and obligate apomictic species. One way of obtaining novel germplasm is by induced mutations. In this work, physical and chemical mutations are combined with in vitro selection procedures seeking for new germplasm, with emphasis on salinity and drought tolerance. Mature seeds of Cenchrus ciliaris L. cv Biloela were subjected to treatments with X rays (400 Gy) and ethyl methanesulphonate (EMS) water solution (5.5 mM for 24 h). To perform in vitro selection, after 7 days of EMS or X More >

A cryopreservation protocol for immature zygotic embryos of species of Ilex (Aquifoliaceae)

LUIS A. MROGINSKI*, PEDRO A. SANSBERRO, ADRIANA M. SCOCCHI, CLAUDIA LUNA, HEBE Y. REY

    BIOCELL, Vol.32, No.1, pp. 33-39, 2008, DOI:10.32604/biocell.2008.32.033

Abstract Tropical Ilex species have recalcitrant seeds. This work describes experiments demonstrating the feasibility of long-term conservation of Ilex brasiliensis, I. brevicuspis, I. dumosa, I. intergerrima, I. paraguariensis, I. pseudoboxus, I. taubertiana, and I. theezans through cryopreservation of zygotic rudimentary embryos at the heart developmental stage. The embryos were aseptically removed from the seeds and precultured (7 days) in the dark, at 27± 2ºC on solidified (0.8% agar) 1/4MS medium, [consisting of quarterstrength salts and vitamins of Murashige and Skoog (1962) medium] with 3% sucrose and 0.1 mg/l Zeatin. The embryos were then encapsulated in 3% calcium alginate beads… More >

Behavior and preservation of an in vitro collection of European aspen in Spain

M.T. MARTIN*, H.E. PEDRANZANI**, R. SIERRA DE GRADO

    BIOCELL, Vol.31, No.1, pp. 41-49, 2007, DOI:10.32604/biocell.2007.31.041

Abstract An in vitro collection has been established with selected European aspen from Palencia province (Spain). Currently, this collection includes 32 high quality clones, selected for their good bearing and healthy state. Most of them belong to different discrete local populations.
Populus tremula L. was propagated in proliferation Aspen Culture Medium; they required subculture every 3 months. The purpose of this study was, therefore, to select a medium which allows the maintenance of 32 clones for a period longer than 3 months without subculture and to observe the behavior of those clones in 15 different culture medium compositions. Seven… More >
