Abstract Long noncoding RNA (lncRNA) taurine-upregulated gene 1 (TUG1) is involved in the development and carcinogenesis of various tumors, suggesting the diagnostic potential of TUG1 in these cancers. However, the exact role of TUG1 and its underlying mechanism in gastric cancer (GC) remain unknown. In this study, the expression of TUG1 and miR-145-5p in GC cell lines and nonmalignant gastric epithelial cell lines was detected by qRT-PCR. BGC-823 and SGC-7901 cells were transfected with si-TUG1, pcDNA 3.1-TUG1, miR-145-5p mimics, or matched controls. The biological function of TUG1 and miR-145-5p in GC cell proliferation and invasion in… More >

An integrated analysis of mRNA-miRNA transcriptome data revealed hub regulatory networks in three genitourinary cancers

    BIOCELL, Vol.41, No.1, pp. 19-26, 2017, DOI:10.32604/biocell.2017.41.019

Abstract Bladder, kidney, prostate and testicular carcinoma are the top four genitourinary cancers in China. Here we analyzed mRNA and miRNA expression profiles of carcinomas of the bladder (TCC), kidney (ccRCC) and testis (TGCT) to uncover their specific regulatory mechanisms. The gene expression profiles of GSE31617 were downloaded from GEO database, which contained 27 samples, including 10 TCC, 7 TGCT and 10 ccRCC. Specific up- and downregulated differentially expressed genes (DEGs) and differentially expressed microRNAs (DEmiRNAs) of each cancer were selected and target genes of DEmiRNAs were predicted. Gene interaction network of the shared genes and… More >

MicroRNA-21 Regulates the Proliferation, Differentiation, and Apoptosis of Human Renal Cell Carcinoma Cells by the mTOR-STAT3 Signaling Pathway

Tao Liang, Xiao-Yong Hu, Yong-Hui Li, Bin-Qiang Tian, Zuo-Wei Li, Qiang Fu

    Oncology Research, Vol.24, No.5, pp. 371-380, 2016, DOI:10.3727/096504016X14685034103356

Abstract MicroRNA-21 (miRNA-21), a kind of short, noncoding RNAs, functioned as a tumor marker and was upregulated in renal cell carcinoma (RCC). However, the underlying mechanisms of miRNA-21 in RCC were uncertain. Therefore, the effects and mechanisms of miRNA-21 on the proliferation, differentiation, and apoptosis of cultured human RCC cells were further investigated in this study. After slicing miRNA-21 in RCC cells, the viability, mRNA expression of C/EBPα and PPARγ, caspase 3 activity, and protein expression of mTOR, STAT3, and pSTAT3 were determined. It was found that knockdown of miRNA-21 downregulated the optical density (OD) value More >

Gemcitabine Plus Vinorelbine as Second-Line Therapy in Patients With Metastatic Esophageal Cancer Previously Treated With Platinum-Based Chemotherapy

    Yue-shen Wang*1, Jing Tian†1, Yong Han, Shu-mei Han, Sheng-bin Shi

    Oncology Research, Vol.24, No.2, pp. 129-135, 2016, DOI:10.3727/096504016X14618564639213

Abstract We evaluated the efficacy and feasibility of the combination of gemcitabine plus vinorelbine in patients with platinum-based chemotherapy-refractory esophageal cancer. We enrolled 35 patients who received gemcitabine plus vinorelbine as second-line treatment after platinum-based chemotherapy failure between May 2009 and April 2012. Dosage: gemcitabine 1,000 mg/m2 plus vinorelbine 25 mg/m2 ; all drugs were administered on days 1 and 8 of a 21-day cycle, and this was continued until failure or unacceptable toxicity. A total of 125 cycles of treatment were administered, and all patients received at least two cycles of treatment (two to five cycles;… More >

miRNA-497 Negatively Regulates the Growth and Motility of Chondrosarcoma Cells by Targeting Cdc25A

Yandong Lu*1, Fangguo Li*1, Tao Xu, Jie Sun*1

    Oncology Research, Vol.23, No.4, pp. 155-163, 2015, DOI:10.3727/096504016X14519157902681

Abstract Chondrosarcoma (CHS) is the second most common malignant bone sarcoma with increased risk of invasion and metastasis. However, the regulatory mechanisms of CHS tumorigenesis remain unknown. Here we investigated the novel role of miR-497 in regulating chondrosarcoma cell growth and cell cycle arrest. RT-PCR analysis showed that the expression of miR-497 is aberrantly downregulated in human chondrosarcoma samples and cells. After transfection with miR-497 mimic or antagomir, the proliferation and apoptosis of JJ012 and OUMS-27 chondrosarcoma cells were determined by CCK-8 assay and flow cytometric analysis, respectively. Results showed that the proliferation capacity of JJ012… More >

BMP-7 Attenuates TGF-β1-Induced Fibronectin Secretion and Apoptosis of NRK-52E Cells by the Suppression of miRNA-21

Zhong Yu, Xu Zai-Chun, Han Wun-Lun, Zhu Yun-Yun

    Oncology Research, Vol.23, No.4, pp. 147-154, 2015, DOI:10.3727/096504016X14519157902645

Abstract Bone morphogenetic protein-7 (BMP-7) inhibited the pathogenesis of renal injury in response to a variety of stimuli. However, little is known about the molecular regulation and mechanism of endogenous BMP-7 and its renoprotective functions. This study examined the regulation of BMP-7 and its role in the fibronectin secretion and apoptosis of NRK-52E cells resulting from transforming growth factor-β1 (TGF-β1) in vitro. Results showed that TGF-β1 promoted factor-associated suicide (FAS), FAS ligand (FASL), fibronectin (FN), and miRNA-21 expression, while it downregulated phospho-Smad1 (pSmad1), pSmad5, and pSmad8 expressions in NRK-52E cells. In contrast, BMP-7 alleviated TGF-β1-induced cell More >
