Abstract Objective: The plastic role of regulatory factor X1 (RFX1) in colon cancer progression and its impact on the tumor microenvironment remain poorly understood. The study aimed to clarify the molecular and clinical role of RFX1 in colon cancer. Methods: We classified colon cancers into subgroups with high and low RFX1 expression and characterized their immune profiles, mutational profiles, cancer immunotherapy and drug sensitivity. By combining RFX1 expression with persistent tumor mutational burden, we proposed a novel nomogram clinical prediction model and validated its predictive performance, and the correlation between high expression and poor prognosis. CMES-Computer Modeling in Engineering & Sciences, Vol.144, No.3, pp. 3585-3605, 2025, DOI:10.32604/cmes.2025.069731 - 30 September 2025

Abstract Lightweight deep learning models are increasingly required in resource-constrained environments such as mobile devices and the Internet of Medical Things (IoMT). Multi-head convolution with channel attention can facilitate learning activations relevant to different kernel sizes within a multi-head convolutional layer. Therefore, this study investigates the capability of novel lightweight models incorporating residual multi-head convolution with channel attention (ResMHCNN) blocks to classify medical images. We introduced three novel lightweight deep learning models (BT-Net, LCC-Net, and BC-Net) utilizing the ResMHCNN block as their backbone. These models were cross-validated and tested on three publicly available medical image datasets:… More >

Abstract Background: Colorectal cancer (CRC) holds the third position in global cancer prevalence mortality. Although chemotherapy is a conventional treatment, recent investigations have shed light on the therapeutic potential of the cGAS cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway in CRC management. Despite the primary role of the death domain-associated protein (Daxx) in cellular apoptosis, its influence on the regulation of cGAS-STING activation remains elusive. Methods: The Daxx degradation and speck formation were conducted using immunofluorescence and Western blotting. The Daxx knock-down and over-expression in CRC cells were performed to detect in vivo and in vitroMore >

Abstract The interest in using the Datura stramonium plant is due to its natural products, which are used in many pharmaceutical industries. The objective of the current study was to assess the therapeutic and cytotoxic effects of the D. stramonium plant on two types of human cancer cell models (MCF7 and HT29) in vitro. A soxhlet apparatus was used to obtain methanolic extract from dried plant leaves. The recovered crude, after the solvent had evaporated, was then dispersed at varied concentrations of extract 100, 50, 20, and 0.0 µg/mL and tested to see how the cells responded. Also,… More >

Abstract Background: Polymethoxylated flavones (PMFs) are compounds present in citrus peels and other Rutaceae plants, which exhibit diverse biological activities, including robust antitumor and antioxidant effects. However, the mechanism of PMFs in reversing drug resistance to colon cancer remains unknown. In the present study, we aimed to investigate the potential connection between the aerobic glycolysis-ROS-autophagy signaling axis and the reversal of PTX resistance in colon cancer by PMFs. Methods: MTT Cell viability assay and colony formation assay were used to investigate the effect of PMFs combined with PTX in reversing HCT8/T cell resistance ex vivo; the mRNA… More > Graphic Abstract

Abstract Cancer poses a significant threat due to its aggressive nature, potential for widespread metastasis, and inherent heterogeneity, which often leads to resistance to chemotherapy. Lung cancer ranks among the most prevalent forms of cancer worldwide, affecting individuals of all genders. Timely and accurate lung cancer detection is critical for improving cancer patients’ treatment outcomes and survival rates. Screening examinations for lung cancer detection, however, frequently fall short of detecting small polyps and cancers. To address these limitations, computer-aided techniques for lung cancer detection prove to be invaluable resources for both healthcare practitioners and patients alike.… More >

Abstract This study investigates the application of deep learning, ensemble learning, metaheuristic optimization, and image processing techniques for detecting lung and colon cancers, aiming to enhance treatment efficacy and improve survival rates. We introduce a metaheuristic-driven two-stage ensemble deep learning model for efficient lung/colon cancer classification. The diagnosis of lung and colon cancers is attempted using several unique indicators by different versions of deep Convolutional Neural Networks (CNNs) in feature extraction and model constructions, and utilizing the power of various Machine Learning (ML) algorithms for final classification. Specifically, we consider different scenarios consisting of two-class colon… More >

Abstract Background: Oleanolic acid (OA), a pentacyclic triterpenoid exhibiting specific anti-cancer properties and highly effective antioxidant activity, was isolated from traditional Chinese medicinal herbs. Conversely, the OA that impacts colon cancer (CC) cells and its underlying mechanisms remain poorly understood. Methods: The cytotoxic effect of OA alone or OA-5-Fluorouracil (5-FU) combination on normal and CC cells was analyzed by methyl thiazolyl diphenyl-tetrazolium bromide (MTT). Then, the impact of OA on CC cell lines (LoVo and HT-29) proliferation and stemness were measured using colon formation and tumorsphere formation assays.
