    BIOCELL, Vol.48, No.2, pp. 283-291, 2024, DOI:10.32604/biocell.2023.045429

Abstract Background: α7 nicotinic acetylcholine receptor (α7nAChR) has been demonstrated to be involved in numerous of inflammatory diseases. Cell pyroptosis is a kind of cell death accompanied by inflammation. Objectives: The objective of this work is to explore the function of α7nAChR on cell pyroptosis in cervical cancer cells. Methods: Immunoblotting, quantitative polymerase chain reaction, and enzyme-linked immunosorbent assay were employed to examine the function of α7nAChR on cell pyroptosis and metabolic changes. Results: Herein, we found that α7nAChR inhibition led to cell pyroptosis in HeLa and SiHa of cervical cancer cells, which was attributed to the upregulation of the polyol… More >

E3 ubiquitin ligase CBL-B suppresses vascular endothelial cell pyroptosis and injury in intracranial aneurysm by facilitating NLRP3 degradation

    WEI ZHENG1, CHENG LIU2,*

    BIOCELL, Vol.48, No.2, pp. 293-301, 2024, DOI:10.32604/biocell.2023.044211

Abstract Objective: Intracranial aneurysm (IA) represents a devastating disease with high rates of disability and mortality, which is initiated by dysfunction of endothelial cells (ECs). Evidence suggests the dysregulation of the E3 ubiquitin ligase family during EC injury. In this work, the role of an E3 ubiquitin ligase, casitas B lymphoma-B (CBL-B), was explored in human brain microvascular EC (HBMEC) function through the NLRP3 pathway. Methods: In vitro IA model was induced by treating HBMECs with oxidized low-density lipoprotein (ox-LDL). The levels of CBL-B and pyroptosis-related proteins NLRP3, ASC, cleaved caspase-1, and GSDME-N were determined by real-time-quantitative polymerase chain reaction and… More >

Suppression of cell pyroptosis by omeprazole through PDE4-mediated autophagy in gastric epithelial cells

    LIPING YE1,2,3,#, HUIYAN SUN4,#, XINHUA LIANG2,#, WENXU PAN2, LI XIANG2,3, WENJUN DU2, LANLAN GENG2, WANFU XU2,3,*, SITANG GONG1,2,3,*

    BIOCELL, Vol.47, No.12, pp. 2709-2719, 2023, DOI:10.32604/biocell.2023.044295

Abstract Introduction: Helicobacter pylori is a risk factor for the development of peptic ulcers with autophagy dysfunction. Omeprazole was widely known as the first-line regimen for H. pylori-associated gastritis. Objectives: The objective of this work was to assess the role of omeprazole on cell pyroptosis and autophagy. Methods: The clinical samples were collected. Quantitative polymerase chain reaction, western blotting, enzyme linked immunosorbent assay, and immunofluorescence (IF) analysis were conducted to reveal the mechanism of omeprazole on cell pyroptosis and autophagy. Results: The results revealed that omeprazole could decrease cell pyroptosis, which was attributed to the downregulation of cleaved caspase-1 expression, resulting… More >

Puerarin inactivates NLRP3-mediated pyroptotic cell death to alleviate cerebral ischemia/reperfusion (I/R) injury through modulating the LncRNA DUXAP8/miR-223-3p axis

ZHENGUO SHI#,*, QIAOYUN WU#, HAIYAN SHI, SONGTIE YING, LIANG TAO

    BIOCELL, Vol.46, No.4, pp. 979-988, 2022, DOI:10.32604/biocell.2022.015345

Abstract NLRP3 inflammasome-mediated cell pyroptosis aggravates the development of cerebral ischemia/reperfusion (I/R) injury, and the aim of this study is to investigate the potential utilization of the Chinese medicine, Puerarin, in treating this disease. Through conducting in vitro and in vivo experiments, the present study illustrated that Puerarin regulated LncRNA double homeobox A pseudogene 8 (DUXAP8)/miR-223-3p axis to inactivate NLRP3-mediated pyroptotic cell death, resulting in the attenuation of I/R injury. Specifically, the cerebral I/R injury in rat models and hypoxia/reoxygenation (H/R) in primary hippocampus neuron (PHN) cells were inducted, which were subsequently exposed to Puerarin treatment. As expected, we validated that… More >
