Abstract With the continuous development of artificial intelligence and machine learning techniques, there have been effective methods supporting the work of dermatologist in the field of skin cancer detection. However, object significant challenges have been presented in accurately segmenting melanomas in dermoscopic images due to the objects that could interfere human observations, such as bubbles and scales. To address these challenges, we propose a dual U-Net network framework for skin melanoma segmentation. In our proposed architecture, we introduce several innovative components that aim to enhance the performance and capabilities of the traditional U-Net. First, we establish… More >

Cell Death of Tumor Melanocytes and Treatment Options

Abstract Melanomas are aggressive cancers, with a high rate of metastatic disease. Cutaneous (CM) and uveal (UM) melanomas are intrinsically different diseases, and most cell death inducers effective for CM do not function for UM. This is primarily due to the fact the eye is an immunologically privileged organ, and it fails to achieve the efficacy of immune checkpoint inhibitors (ICIs) comparable to that for CM. However, approaches utilizing specific melanoma-associated antigens are being developed for metastatic forms of CM and UM. The most promising to date are gp100 and tyrosinase related protein 1 (TYRP1), primarily… More >

Abstract Glutamine metabolism (GM) plays an important role in tumor growth and proliferation. Skin cutaneous melanoma (SKCM) is a glutamine-dependent cancer. However, the molecular characteristics and action mechanism of GM on SKCM remain unclear. Therefore, we aimed to explore the effects of GM-related genes on survival, clinicopathological characteristics, and the tumor microenvironment in SKCM. In this study, 682 SKCM samples were obtained from the Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. Consensus clustering was used to classify SKCM samples into distinct subtypes based on 41 GM-related genes. Differences in survival, immune infiltration, clinical… More > Graphic Abstract

Abstract Objective: Stage II melanoma patients have high risk for regional and distant metastases and may benefit from novel therapeutic strategies. To clarify the role of NK cells in Stage II melanoma, we characterized the cytotoxic activity of NK cells and the expression of various activating and inhibitory receptors in high-risk cutaneous melanoma patients (Stages IIB and IIC) compared to low-risk patients (Stage IA). Materials and Methods: Native and cytokinetreated peripheral blood mononuclear cells were used for functional and phenotypical analyses. Results: Compared to Stage IA-B patients, Stage IIB-C patients showed significantly decreased NK cell activity,… More >
