Changes of protein expression during tumorosphere formation of small cell lung cancer circulating tumor cells

    SANDRA STICKLER1, BARBARA RATH1, MAXIMILIAN HOCHMAIR2, CLEMENS LANG3, LUKAS WEIGL4, GERHARD HAMILTON1,*

    Oncology Research, Vol.31, No.1, pp. 13-22, 2023, DOI:10.32604/or.2022.027281

Abstract Small cell lung cancer (SCLC) is frequently disseminated and has a dismal prognosis with survival times of approximately two years. This cancer responds well to initial chemotherapy but recurs within a short time as a globally chemoresistant tumor. Circulating tumor cells (CTCs) are held responsible for metastasis, the extremely high numbers of these cells in advanced SCLC allowed us to establish several permanent CTC cell lines. These CTCs are distinguished by the spontaneous formation of large spheroids, termed tumorospheres, in regular tissue culture. These contain quiescent and hypoxic cells in their interior and are associated with high chemoresistance compared to… More >

Ectopic Expression of miR-147 Inhibits Stem Cell Marker and Epithelial–Mesenchymal Transition (EMT)-Related Protein Expression in Colon Cancer Cells

Xiaofei Ning*, Cong Wang, Meng Zhang, Kecheng Wang*

    Oncology Research, Vol.27, No.4, pp. 399-406, 2019, DOI:10.3727/096504018X15179675206495

Abstract Colon cancer is one of the most common cancers in the world. Epithelial-to-mesenchymal transition (EMT) is a crucial step in tumor progression and is also involved in the acquisition of stem cell-like properties. Some miRNAs have been shown to function as either tumor suppressors or oncogenes in colon cancer. Here we investigated the role of miR-147 in the regulation of the stem cell-like traits of colon cancer cells. We observed that miR-147 was downregulated in several colon cancer cell lines, and overexpressed miR-147 decreased the expression of cancer stem cell (CSC) markers OCT4, SOX2, and NANOG in the colon cancer… More >

YB-1 downregulation attenuates UQCRC1 protein expression level in H9C2 cells and decreases the mitochondrial membrane potential

    HUIFANG CHEN1,2, XIAOYING ZHOU2, ZONGHONG LONG2, XIANGLONG TANG2, HONG LI2,*

    BIOCELL, Vol.44, No.3, pp. 371-379, 2020, DOI:10.32604/biocell.2020.08893

Abstract UQCRC1 is one of the 10 mitochondrial complex III subunits, this protein has a role in energy metabolism, myocardial protection, and neurological diseases. The upstream mechanism of the UQCRC1 protective effect on cardiomyocytes is currently unavailable. In order to explore the upstream molecules of UQCRC1 and elucidate the protective mechanism of UQCRC1 on cardiomyocytes in more detail, we focused on the nuclease-sensitive elementbinding protein 1 (YB-1). We hypothesized YB-1 acts as an upstream regulatory molecule of UQCRC1. This study found that YB-1 RNAi significantly reduces the expression of the UQCRC1 protein level (p < 0.05) and obviously decreases the mitochondrial… More >

Brief Note: Influence of 2-methoxyestradiol on MCF-7 cells: An improved differential interference contrasting technique and Bcl-2 and Bax protein expression levels

    ANNIE JOUBERT1*, SUMARI MARAIS1, CHRISTINE MARITZ2

    BIOCELL, Vol.33, No.1, pp. 67-70, 2009, DOI:10.32604/biocell.2009.33.067

Abstract Proteins of the B-cell lymphoma 2 family are crucial for the regulation of apoptosis. B-cell lymphoma 2-associated X is a pro-apoptotic protein, while B-cell lymphoma 2 protein opposes apoptosis. The influence of 1 μM 2-methoxyestradiol was investigated on the expression levels of these two proteins in MCF-7 cells. 2-Methoxyestradiol exposure did not influence B-cell lymphoma 2 protein expression levels after 24 h of exposure. In contrast, B-cell lymphoma 2-associated X protein levels were significantly reduced. An improved differential interference contrasting technique revealed compromised cell density and the presence of a mitotic block in exposed cells. The study proposes that the… More >

Abstract Progenitor cells play an important biological role in tooth and bone formation, and previous analyses during bone and dentine induction have indicated that they may be a good alternative for tissue engineering. Thus, to clarify the influence of the microenvironment on protein and gene expression, MDPC23 cells (mouse dental papilla cell line) and KUSA/A1 cells (bone marrow stromal cell line) were used, both in vitro cell culture and in intra-abdominal diffusion chambers implanted in 4-week-old male immunodefficient mice (SCID mice). Our results indicate that KUSA/A1 cells differentiated into osteoblast-like cells and induced bone tissue inside the chamber, whereas, MDPC-23 showed… More >

Physiological Processes Contributing to the Difference in Grain Amino Acid Content between Two Hybrid Rice (Oryza sativa L.) Cultivars

Min Huang*, Hengdong Zhang, Chunrong Zhao, Guanghui Chen and Yingbin Zou

    Phyton-International Journal of Experimental Botany, Vol.88, No.2, pp. 101-107, 2019, DOI:10.32604/phyton.2019.06694

Abstract Improving grain amino acid content of rice (Oryza sativa L.) is essential for the health of consumers. This study was conducted to identify the physiological processes that contribute to the higher grain amino acid content in hybrid rice cultivar Lingliangyou 268 compared to Luliangyou 996. The results showed that total amino acid content in grains was 9% higher in Lingliangyou 268 than in Luliangyou 996. There was no significant difference in grain nitrogen (N) content between Lingliangyou 268 and Luliangyou 996, while ratio of amino acid to N was 6% higher in Lingliangyou 268 compared to Luliangyou 996. A total… More >
