Abstract Gastric cancer is one of the most common malignancies. DNA methylation is implicated in DNA mismatch repair genes deficiency. In the present study, we evaluated the methylation status of MLH1, MSH2, MSH6 and PMS2 in 20 diffuse- and 26 intestinal-type gastric cancer samples and 20 normal gastric mucosal of gastric cancer patients from Northern Brazil. We found that none of the nonneoplastic samples showed methylation of any gene promoter and 50% of gastric cancer samples showed at least one methylated gene promoter. Methylation frequencies of MLH1, MSH2, MSH6 and PMS2 promoter were 21.74%, 17.39%, 0% and 28.26% respectively in gastric… More >

Abstract Glioma is one of the lethal central nervous system tumors. The infiltrative and invasive growth nature makes it difficult to identify the boundary between glioma and the normal tissues, resulting in inevitable recurrence after surgery operation. Gliomas do not metastasize, so to prevent the residual tumor from proliferating or invading is a key challenge. Previous report indicated that hypotaurine could facilitate glioma invasion and suppress demethylases’ activities. Using a hypotaurine synthesis deficient U251 cell line, we proved that the cells invasion ability was impaired. Gene expression profile analysis exhibited that knocking down one of the key enzymes of hypotaurine synthesis,… More >

Abstract Today, prognosis, diagnosis and treatment of cancers are progressing with non-invasive methods, including investigation and modification of the DNA methylation profile in cancer cells. One of the effective factors in regulating gene expression in mammals is DNA methylation. Methylation alterations of genes by external factors can change the expression of genes and inhibit the cancer. In the present study, we investigated the effect of Down syndrome critical region 1 gene (DSCR1) ectopic expression on the methylation status of the BCL-XL, ITGA6, TCF3, RASSF1A, DOK7, VIM and CXCR4 genes in breast cancer cell lines. The effect of DSCR1 ectopic expression on… More >
