Abstract Introduction: Dexamethasone (Dex) caused impaired osteoblast differentiation and oxidative stress (OS) in bone marrow mesenchymal stem cells (BMSCs). This work sought to elucidate the precise molecular pathway through which Dex influences osteogenic differentiation (OD) and OS in BMSCs. Methods: The expression of Runt-related transcription factor 1 (RUNX1) and alpha-2 macroglobulin (A2M) was assessed in Dex-treated BMSCs using qRT-PCR and Western Blot. Following the functional rescue experiments, cell proliferation was determined by MTT assay, reactive oxygen species (ROS) expression by DCFH-DA fluorescent probe, lactate dehydrogenase (LDH), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (Gpx) expression by kits, OD by alkaline… More >

Abstract Castor is one of the top 10 oil crops in the world and has extremely valuable uses. Castor inflorescences directly affect yield, so the study of inflorescence development is very important in increasing castor yield. Our previous studies have shown that the PIP5K gene family (PIP5Ks) is associated with inflorescence development. In this study, to determine the function of each PIP5K gene in castor, a female Lm-type castor line, aLmAB2, was used to determine the relative expression levels of the PIP5Ks in castor inflorescences. Six PIP5K genes were heterologously overexpressed in Arabidopsis thaliana, the relative expression of each gene and… More >

    Phyton-International Journal of Experimental Botany, Vol.92, No.11, pp. 3063-3075, 2023, DOI:10.32604/phyton.2023.043803

Abstract Adventitious root formation poses a major constraint on the tissue culture and genetic transformation of Eucommia ulmoides. Micrografting offers a new method for the transplantation of genetic transformation, and its success depends on the formation of graft unions. This study used transgenic rootless test-tube seedlings as scions and seedlings from seed as rootstocks during micrografting to avoid the rooting issues that occur during tissue culture and to investigate the role of the EuEG1 gene in the graft healing process. We found that the EuEG1 gene is a vital regulator of graft, and its overexpression contributes to the survival of Eucommia… More >

Abstract The auxin polar transporter, PIN-FORMED 2 (PIN2) plays an important role in root development. However, it remains unclear whether PIN2 genes form two Liriodendron species, L. chinense (LcPIN2) and L. tulipifera (LtPIN2), are both involved in root development and whether and to what extent these two genes diverge in function. Here, we cloned and overexpressed LcPIN2 and LtPIN2 in Arabidopsis thaliana wild-type (WT) and Atpin2 mutant. Phylogenetic and sequence analysis showed a small degree of differentiation between these two Liriodendron PIN2 genes. Tissue-specific gene expression analysis indicated that both Liriodendron PIN2 genes were highly expressed in roots, implying a potential… More >

Abstract EARLY FLOWERING 3 (ELF3), a light zeitnehmer (time-taker) gene, regulates circadian rhythm and photoperiodic flowering in Arabidopsis, rice, and barley. The three orthologs of ELF3 (TaELF3-1AL, TaELF3-1BL, and TaELF3-1DL) have been identified in wheat too, and one gene, TaELF3-1DL, has been associated with heading date. However, the basic characteristics of these three genes and the roles of the other two genes, TaELF3-1BL and, TaELF3-1AL, remain unknown. Therefore, the present study obtained the coding sequences of the three orthologs (TaELF3-1AL, TaELF3-1BL, and TaELF3-1DL) of ELF3 from bread wheat and characterized them and investigated the role of TaELF3-1BL in Arabidopsis. Protein sequence… More >

    Oncology Research, Vol.29, No.4, pp. 291-303, 2021, DOI:10.32604/or.2022.025172

Abstract Increasing numbers of long noncoding RNAs (lncRNAs) are implicated in breast cancer oncogenicity. However, the contribution of LINC02568 toward breast cancer progression remains unclear and requires further investigation. Herein, we evaluated LINC02568 expression in breast cancer and clarified its effect on disease malignancy. We also investigated the mechanisms underlying the pro-oncogenic role of LINC02568. Consequently, LINC02568 was upregulated in breast cancer samples, with a notable association with worse overall survival. Functionally, depleted LINC02568 suppressed cell proliferation, colony formation, and metastasis, whereas LINC02568 overexpression exerted the opposite effects. Our mechanistic investigations suggested that LINC02568 was physically bound to and sequestered microRNA-874-3p… More >
