The SS18-SSX fusion oncoprotein: Friend and foe in targeted therapy for synovial sarcoma

GAVIN M. ANCHONDO, KYRA PARKER, ALEXIS BRUCE, ELIZABETH CORTEZ, LE SU*

    Oncology Research, Vol.33, No.5, pp. 1001-1005, 2025, DOI:10.32604/or.2025.060573 - 18 April 2025

Abstract Synovial sarcoma is a high-grade soft tissue malignancy characterized by a unique fusion gene known as SS18-SSX. The SS18-SSX fusion protein acts as an oncogenic driver of synovial sarcoma, and it has thus been commonly accepted that disruption of SS18-SSX function represents a therapeutic means of treating synovial sarcoma, but emerging evidence suggests that upon depletion of SS18-SSX, an anti-apoptotic signal surprisingly arises to protect synovial sarcoma cell survival. In this article, we discuss the controversial roles of SS18-SSX’s transcriptional activity in synovial sarcoma biology and outline a synergistic strategy for overcoming the resistance of More > Graphic Abstract

Multiple Recurrence of Primary Orbital Synovial Sarcoma: Report of Two Cases and Literature Review

Yi Wu, Yujiao Wang, Weimin He*

    Oncologie, Vol.24, No.4, pp. 927-935, 2022, DOI:10.32604/oncologie.2022.026720 - 31 December 2022

Abstract Synovial sarcoma (SS) is typically an aggressive malignant soft tissue tumor that mostly affects adolescents and young adults. It is extremely rare in orbit and carries a high risk of recurrence and metastasis, posing a challenge to ophthalmologists in diagnosing and managing. We present two primary orbital synovial sarcoma cases with unilateral exophthalmos and limited motility. Both male patients underwent reoperation in our hospital since tumor recurrence; the pathologic diagnoses were biphasic type and occult type, respectively. Both cases were positive for EMA and CK, and SOX-9 and INI-1 were newly discovered immune markers. Fluorescence in More >

    P. Arunachalam1, N. Janakiraman1, Junaid Rashid2, Jungeun Kim2,*, Sovan Samanta3, Usman Naseem4, Arun Kumar Sivaraman5, A. Balasundaram6

Abstract In this research work, we proposed a medical image analysis framework with two separate releases whether or not Synovial Sarcoma (SS) is the cell structure for cancer. Within this framework the histopathology images are decomposed into a third-level sub-band using a two-dimensional Discrete Wavelet Transform. Subsequently, the structure features (SFs) such as Principal Components Analysis (PCA), Independent Components Analysis (ICA) and Linear Discriminant Analysis (LDA) were extracted from this sub-band image representation with the distribution of wavelet coefficients. These SFs are used as inputs of the Support Vector Machine (SVM) classifier. Also, classification of PCA… More >

Synovial Sarcoma Classification Technique Using Support Vector Machine and Structure Features

    P. Arunachalam1, N. Janakiraman1,*, Arun Kumar Sivaraman2, A. Balasundaram3, Rajiv Vincent2, Sita Rani4, Barnali Dey5, A. Muralidhar2, M. Rajesh2

    Intelligent Automation & Soft Computing, Vol.32, No.2, pp. 1241-1259, 2022, DOI:10.32604/iasc.2022.022573 - 17 November 2021

Abstract Digital clinical histopathology technique is used for accurately diagnosing cancer cells and achieving optimal results using Internet of Things (IoT) and blockchain technology. The cell pattern of Synovial Sarcoma (SS) cancer images always appeared as spindle shaped cell (SSC) structures. Identifying the SSC and its prognostic indicator are very crucial problems for computer aided diagnosis, especially in healthcare industry applications. A constructive framework has been proposed for the classification of SSC feature components using Support Vector Machine (SVM) with the assistance of relevant Support Vectors (SVs). This framework used the SS images, and it has… More >

    Canadian Journal of Urology, Vol.20, No.2, pp. 6734-6736, 2013

Abstract We present a case report of a 17-year-old patient with a large renal mass that was detected on a computed tomography scan during investigation for secondary hypertension. Radical nephrectomy was performed, and the morphologic and immunocytochemical findings were compatible with a diagnosis of monophasic synovial sarcoma of the kidney. A cytogenetic search for t(X;18) translocation was performed, which was negative. The patient underwent an ifosfamide-based chemotherapy regimen. During follow-up, a positron emission tomography scan showed increased 18F-fluorodeoxyglucose metabolism at the right femur. Although cancer cells were expected in the biopsy specimen, only fibrous dysplasia of More >
