    Phyton-International Journal of Experimental Botany, Vol.91, No.1, pp. 83-96, 2022, DOI:10.32604/phyton.2022.017281

Abstract In Mexico, there is a need to produce large quantities of plantlets for the establishment and replanting of blue (cv. azul) agave production areas. Most of these plots are within the origin denomination area (DOT, Spanish acronym) of the distilled product of this plant, known as tequila. The objective of this study was to develop an in vitro-propagation protocol for Agave tequilana Weber cv. azul using segmented stems in both: solid and liquid media. A disinfection and in vitro technique were developed to obtain shoots, through plantlets collected in commercial plots, which attained 100% surface-disinfection and budding rate. At the… More >

Influence of genotype and explant source on indirect organogenesis by in vitro culture of leaves of Melia azedarach L.

    S.K. VILA*, H.Y. REY, L.A. MROGINSKI

    BIOCELL, Vol.28, No.1, pp. 35-41, 2004, DOI:10.32604/biocell.2004.28.035

Abstract In vitro regeneration of shoots from leaf explants of the Paradise tree (Melia azedarach L.) was studied. Three different portions (proximal portion, distal portion and rachis of the leaflets) of three developmental stages (folded, young still expanding and completely expanded) of leaves of 10 – 15 years old plants of seven genotypes were cultured on Murashige and Skoog (1962) medium (MS) supplemented with 1mg.l-1 benzylaminopurine (BAP) + 0.1mg.l-1 kinetin (KIN) + 3 mg.l-1 adenine sulphate (ADS).
The rachis of the leaflets of the completely expanded leaves was found to be the most responsive tissue, in most of the genotypes… More >

Micropropagation of wild grapevines (Vitis spp.) of Central Mexico

    Jiménez-Martínez JH1, MG Gutiérrez-Martínez2, O Franco-Mora1, A González-Huerta2, AT Gutiérrez-Ibáñez2

    Phyton-International Journal of Experimental Botany, Vol.82, pp. 107-112, 2013, DOI:10.32604/phyton.2013.82.107

Abstract Two accessions of wild grapevines (Vitis spp.) native to Puebla State, Nealticán and Teziutlán, and other two native to Temascaltepec (1 and 2), Mexico State, were cultured in vitro using both Murashige-Skoog (MS) and woody plant medium (WPM) mediums. There were no differences between mediums for shoot generation. However, differences were shown among accessions for explant vigour, callus development, number of roots, leaves and nodes per explant. Rooting was evaluated only in MS medium by adding naphthalene acetic acid (NAA), indole butyric acid (IBA) or indole acetic acid (IAA) at 0.5 mg/L. With the exception of Temascaltepec 2, NAA generated… More >

Plant micropropagation in experimental lines of forage maize coming from crossing Zea mays L. and Zea diploperennis Iltis, Doebley and Guzmán

Torroba MC, HA Paccapelo, L Aguilera, J Mazzola

    Phyton-International Journal of Experimental Botany, Vol.77, pp. 93-102, 2008, DOI:10.32604/phyton.2008.77.093

Abstract An optimized methodology for improving regeneration of maize plants by direct organogenesis was evaluated. Our objective was to embarobtain genetically homogeneous plants through in vitro methods to regenerate clumps of multiple shoots from shoot tips at high frequency. Cultures were initiated from shoot tips of experimental lines of maize on a Murashige & Skoog (MS) medium supplemented with 2 mg/l benziladenine. Two experimental forage maize lines were used, in which two frequencies of subcultures were evaluated, namely line L. 850 (every 15 or 30 days) and line L.769 (every 30 days). It was observed that the highest rate of regeneration… More >

Stability of protein patterns in coffee seedlings regenerated by somatic embryogenesis

    Menéndez-Yuffá1 A, L Ríos-Bolívar2

    Phyton-International Journal of Experimental Botany, Vol.77, pp. 49-64, 2008, DOI:10.32604/phyton.2008.77.049

Abstract Regeneration and multiplication of coffee (Coffea arabica L.) through somatic embryogenesis can be efficiently achieved through different methods. However, it is very important to analyze the progenies obtained by these methods in order to establish their genetic stability. The objectives of this research were (1) to determine the protein electrophoretic patterns in leaves of coffee vitroplants which were regenerated through somatic embryogenesis, and (2) to compare these patterns with those of the explant donor plants. The protein patterns of some coffee genotypes were initially determined to detect possible differences among them. Each analyzed genotype showed a characteristic set of protein… More >

Brief Note : Improved in vitro embryo development of stenospermic grape by putrescine

MARÍA TERESA PONCE, MÓNICA ELIZABETH GUIÑAZÚ, RICARDO TIZIO

    BIOCELL, Vol.26, No.2, pp. 263-266, 2002, DOI:10.32604/biocell.2002.26.263

Abstract The goal of this study was to determine the effect of putrescine, added to the culture medium, on the in vitro development of stenospermic grape (Vitis vinifera L) embryos. The cross breedings of Perlón x G.C88552 and Perlón x Argentina were used. 0 (control), 2 and 4 mM of putrescine were added to the immature seed’s culture medium. In Perlón x Argentina, 2mM of putrescine statistically increased the percentage of total embryos, direct germination, polyembryos and normal plants. In Perlón x G.C88552, only 2 mM of putrescine increased all the variables considered, eventually tripling the percentage of normal plants obtained.… More >
