Abstract Birch (Betula platyphylla Suk.), distributed in Eurasia, North America, and Australia, is a kind of cold-resistant, fast-growing, and vital pulpwood tree species. It is also one of the most important ecological restoration tree species with high values of economic benefits in Northeast China. To improve the genetic gain and expand the economic benefit of B. platyphylla, many genetic improvements have been carried out. In China, B. platyphylla is widely distributed and varied, and there are many varieties with excellent genetic characteristics. In this paper, the genetic improvement of B. platyphylla was reviewed, and the previous research results were discussed from… More >

    Shikai Zhang1,2, Wang Zhan3, Heng Zhang1,2, Shuanglin Song3, Xibin Qu3, Jiayi Wang3, Mulualem Tigabu4, Xiang Li1,2, Qinhui Zhang1,2, Minghui Zhao1,2, Huanzhen Liu2,*, Xiyang Zhao1,2,*

    Phyton-International Journal of Experimental Botany, Vol.91, No.3, pp. 489-505, 2022, DOI:10.32604/phyton.2022.018321

Abstract Juglans mandshurica is an economically and ecologically valuable species that is used for various construction purposes, making luxurious furniture, as food and sources of medicinal substances and landscaping because of its excellent wood, edible fruits and rich in various types of chemical compounds. In the past few decades, several genetic improvements of J. mandshurica were made, with a focus on the selection of improved varieties and on breeding technology. Many elite provenances and families were selected based on growth traits or wood properties. In recent years, with the increasing demand for high-quality seedlings in Chinese forestry production, the breeding goals… More >

Transcriptome based high-throughput SSRs and SNPs discovery in the medicinal plant Lagenaria siceraria

HONGYUAN ZHANG, MIN ZHANG, JIE TAN, SHUPING HUANG, GUOLIN ZHOU*, XIA CHEN*

    BIOCELL, Vol.45, No.2, pp. 371-386, 2021, DOI:10.32604/biocell.2021.013869

Abstract Lagenaria siceraria (Molina) Standley has unique biological characteristics with high nutritional and medicinal values. It is an important pharmaceutical plant with various biologically active ingredients. Genetic improvement and deeper genomic studies require a rich resource of molecular markers. The application of next-generation sequencing technology, especially for transcriptome profiling, has greatly facilitated high throughput single nucleotide polymorphism (SNP) and simple sequence repeat (SSR) discovery. In this study, we sequenced the transcriptome of three major cultivars of L. siceraria and obtained 64.88 GB of clean data. The assembled high-quality reads were clustered into 89,347 unigenes, which were annotated by non-redundant protein database,… More >

Applications of Molecular Markers in Fruit Crops for Breeding Programs—A Review

Riaz Ahmad1, Muhammad Akbar Anjum1,*, Safina Naz1, Rashad Mukhtar Balal2

    Phyton-International Journal of Experimental Botany, Vol.90, No.1, pp. 17-34, 2021, DOI:10.32604/phyton.2020.011680

Abstract Selection and use of molecular markers for evaluation of DNA polymorphism in plants are couple of the most important approaches in the field of molecular genetics. The assessment of genetic diversity using morphological markers is not sufficient due to little differentiating traits among the species, genera or their individuals. Morphological markers are not only highly influenced by environmental factors but skilled assessment is also prerequisite to find the variations in plant genetic resources. Therefore, molecular markers are considered as efficient tools for detailed DNA based characterization of fruit crops. Molecular markers provide new directions to the efforts of plant breeders… More >

Obtaining new germplasm in Cenchrus ciliaris L. through induced-mutation and in vitro selection

    López Colomba E1, A Prina2, S Griffa1, AN Ribotta1, E Carloni1, E Tommasino1, C Luna1, E Biderbost1, K Grunberg1

    Phyton-International Journal of Experimental Botany, Vol.80, pp. 59-64, 2011, DOI:10.32604/phyton.2011.80.059

Abstract Cenchrus ciliaris L., a forage grass of wide distribution in the north-west of Argentina, is a tetraploid (4x = 36) and obligate apomictic species. One way of obtaining novel germplasm is by induced mutations. In this work, physical and chemical mutations are combined with in vitro selection procedures seeking for new germplasm, with emphasis on salinity and drought tolerance. Mature seeds of Cenchrus ciliaris L. cv Biloela were subjected to treatments with X rays (400 Gy) and ethyl methanesulphonate (EMS) water solution (5.5 mM for 24 h). To perform in vitro selection, after 7 days of EMS or X rays… More >

An efficient protocol for culturing meristems of sorghum hybrids

    Sadia1,3 B, PC Josekutty2, SD Potlakayala2, P Patel2, S Goldman3, SV Rudrabhatla2

    Phyton-International Journal of Experimental Botany, Vol.79, pp. 177-181, 2010, DOI:10.32604/phyton.2010.79.177

Abstract A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. The highest number of shoots per meristem (68 ± 2) was recorded for the genotype… More >

Genetic variability in 22 sugarcane hybrid varieties (Saccharum spp. Hybrid)

Alejandre Rosas1 JA, ME Galindo Tovar2, HE Lee Espinosa2, OG Alvarado Gómez3

    Phyton-International Journal of Experimental Botany, Vol.79, pp. 87-94, 2010, DOI:10.32604/phyton.2010.79.087

Abstract Development of new sugarcane varieties allows to increase sucrose production and faces challenges as new pests and diseases, and soil fertility losses. The objective of this study was to contribute to expand molecular tools in selecting useful progenitors for hybridization programs. With this purpose, 14 RAPDs primers were pre-screened, and four of them which produced clearly resolved, reproducible and polymorphic fragment patterns were selected for genetic analysis of 22 sugarcane variety hybrids. These were used in the Estación de Hibridación at Tapachula, Chiapas, as progenitors in genetic improvement programs. Genetic distance analysis showed three main clusters. The first cluster contained… More >
