Landscape of Sequence Variations in Homologous Copies of FAD2 and FAD3 in Rapeseed (Brassica napus L.) Germplasm with High/Low Linolenic Acid Trait

Haoxue Wu#, Xiaohan Zhang§,#, Xiaoyu Chen, Kang Li, Aixia Xu, Zhen Huang, Jungang Dong, Chengyu Yu*

    Phyton-International Journal of Experimental Botany, Vol.93, No.3, pp. 627-640, 2024, DOI:10.32604/phyton.2024.050321

Abstract Genetic manipulation (either restraint or enhancement) of the biosynthesis pathway of α-linolenic acid (ALA) in seed oil is an important goal in Brassica napus breeding. B. napus is a tetraploid plant whose genome often harbors four and six homologous copies, respectively, of the two fatty acid desaturases FAD2 and FAD3, which control the last two steps of ALA biosynthesis during seed oil accumulation. In this study, we compared their promoters, coding sequences, and expression levels in three high-ALA inbred lines 2006L, R8Q10, and YH25005, a low-ALA line A28, a low-ALA/high-oleic-acid accession SW, and the wildtype ZS11. The expression levels of… More >

Histological Assessment of Bone Regeneration in the Maxilla with Homologous Bone Graft: A Feasible Option for Maxillary Bone Reconstruction

Sergio Henrique Gonçalves Motta1, Ana Paula Ramos Soares1, Juliana Campos Hasse Fernandes2, Gustavo Vicentis Oliveira Fernandes2,3,*

    Journal of Renewable Materials, Vol.12, No.1, pp. 131-148, 2024, DOI:10.32604/jrm.2023.043940

Abstract Bone biomaterials have been increasingly used to reconstruct maxillary atrophic ridges. Thus, the aim of this study was to evaluate bone reconstruction in the maxilla using a homologous cortico-cancellous FFB (lyophilized) graft and verify its reliability. Eight individuals were included from 2014 to 2018. The first surgery was performed to install homologous bone blocks in the maxilla. The period of the second intervention varied between 5 months and 15 days to 11 months (≈7.93 months). The biopsies were taken from the central region of the matured graft during the surgery for implant placement. All patients presented clinical and radiographic conditions… More >

DNA polymerase θ (POLQ): A druggable DNA polymerase for homologous recombination-deficient cancer cells

    MERAN KESHAWA EDIRIWEERA*

    BIOCELL, Vol.47, No.3, pp. 441-444, 2023, DOI: 10.32604/biocell.2023.025747

Abstract Irregularities in the DNA repair pathways are frequently observed in cancer. Dysregulated DNA repair pathways support growth advantages to tumor cells. DNA polymerase-theta (POLQ) is an error-prone DNA polymerase involved in double-strand break repair through microhomology-mediated end joining (MMEJ). POLQ also mediates translesion DNA synthesis and it is largely not expressed in normal cells. POLQ is overexpressed in a range of cancer cells, including homologous recombination (HR) deficient cancer cells. In HR deficient cells, MMEJ is crucial as a backup DNA repair pathway, indicating the indispensable role of POLQ-mediated MMEJ in HR deficient cancer cells. In addition, POLQ is synthetic… More >

    BIOCELL, Vol.30, No.2, pp. 269-278, 2006, DOI:10.32604/biocell.2006.30.269

Abstract Objective: To investigate the functions of Fibroblast Growth Factor Receptor-2 (FGFR2) at different stages of cell differentiation. The engineered murine embryonic stem (ES) cells with conditional knockout of FGFR2 were developed depending on Cre-loxP. Methods: Cre-loxP system was used in a conditional targeting vector. The competent AM-1 bacteria, which expressed Cre-recombinase, was used to confirm the Cre-mediated deletion of the floxed exons 7 and 8 of FGFR2. The targeting vector was electroporated into the ES cells, and the transfected ES cells were screened with G418 and Ganciclovir. Finally, the ES clones with correct targeting events were identified by Southern Blot… More >

    BIOCELL, Vol.30, No.1, pp. 43-49, 2006, DOI:10.32604/biocell.2006.30.043

Abstract Electrophysiological events occur early after fertilization, along with changes in intracellular Ca2+ concentration . Passive electrical parameters were determined in golden hamster oocytes by whole cell patch-clamp method. In separate experiments the effect of 4-aminopyridine on resting oocytes was tested. The single-channel patch clamp configuration was employed to assess the electrical response to fertilization with homologous sperm. Structure of oocytes submitted to patch clamp was evaluated with scanning electron microscopy and found to be preserved.
Oocyte diameter was 70.2 ± 2.2 µm; their resting parameters were: membrane potential 23.8 ± 0.8 mV; total membrane specific resistance 519.1 ± 94.6… More >

Chromosomal location of four genes encoding Class III peroxidases in wheat

    Simonetti E1, E Alba2, A Delibes2

    Phyton-International Journal of Experimental Botany, Vol.81, pp. 139-142, 2012, DOI:10.32604/phyton.2012.81.139

Abstract In a previous work, deduced amino acid sequences from twenty wheat peroxidase genes were assigned to seven groups designated as TaPrx108 to TaPrx114. Some of these apoplastic peroxidases have previously shown to play different roles in the plant defense responses to infection by the cereal cyst nematode Heterodera avenae. In the present study, PCR marker analysis using Sears’s aneuploid wheat lines cv. ‘Chinese Spring’ was used to locate four genes encoding peroxidase isozymes. The TaPrx111-A, TaPrx112-D and TaPrx113-F genes were located on the short arm of chromosome 2B and the TaPrx109-C on the long arm of chromosome 1B. These results… More >

Interaction of bifidobacteria with the gut and their influence in the immune function

Gabriela Perdigón1,2, Mónica Locascio1, Marta Medici1, Aida Pesce de Ruiz Holagado1,2, Guillermo Oliver1

    BIOCELL, Vol.27, No.1, pp. 1-9, 2003, DOI:10.32604/biocell.2003.27.001

Abstract Bifidobacteria are predominant in the lumen of the large intestine and confer various health benefits on the host. They are also used in the preparation of new fermented milks (bioyogurts) or added to conventional yogurt to generate probiotic effects. The colonization of the gut by bacteria tends to be host specific due partly to the way in which bacteria adhere to the intestinal wall. Using a homologous strain of Bifidobacterium animalis in an experimental mouse model, we analyzed by immunofluorescence labelledbacteria and transmission electronic microscopy the importance of this bacterial interaction with epithelial an immune cells associated to the gut,… More >
