    Yong Zhao2,#, Lili Li1,#, Rui Luo1, Mu Peng1, Tongtong Jiang1, Mingda Yin1, Yanpeng Wen1, Zihan Wang1, Fenglan Huang1,4,5,6,7,*, Fanjuan Meng3,*

    Phyton-International Journal of Experimental Botany, Vol.92, No.12, pp. 3237-3250, 2023, DOI:10.32604/phyton.2023.031495

Abstract Castor, scientifically known as Ricinus communis L., is among the top ten oil crops globally. It is considered a renewable resource and is commonly referred to as ‘green oil’. Castor seeds contain castor oil as their main component, which is predominantly composed of ricinoleic acid. This study utilized RNAi technology to silence the NPC6 gene in NO.2129 castor, resulting in the creation of mutant plants L1 and L2. The weight of 100 dry seed kernels from L1 and L2 exceeds that from NO.2129. The crude fat and ricinoleic acid levels of L1 and L2 were higher than those of NO.2129… More >

    Phyton-International Journal of Experimental Botany, Vol.92, No.2, pp. 501-519, 2023, DOI:10.32604/phyton.2022.023479

Abstract High yield, high quality, stable yield, adaptability to growth period, and modern mechanization are the basic requirements for crops in the 21st century. Soybean oleic acid is a natural unsaturated fatty acid with strong antioxidant properties and stability. Known as a safe fatty acid, it has the ability to successfully prevent cardiovascular and cerebrovascular disorders. Improving the fatty acid composition of soybean seeds, can not only speed up the breeding process of high-quality high-oil and high-oleic soybeans, but also have important significance in human health, and provide the possibility for the development of soybean oil as a new energy source.… More >

Construction and Functional Analysis of CRISPR/Cas9 Vector of FAD2 Gene Family in Soybean

Zhifeng Xiao, Yukun Jin, Qi Zhang, Abraham Lamboro, Baozhu Dong, Zhuoyu Yang, Piwu Wang*

    Phyton-International Journal of Experimental Botany, Vol.91, No.2, pp. 349-361, 2022, DOI:10.32604/phyton.2022.017451

Abstract Soybean oleic acid content is one of the important indexes to evaluate the quality of soybean oil. In the synthesis pathway of soybean fatty acids, the FAD2 gene family is the key gene that regulates the production of linoleic acid from soybean oleic acid. In this study, CRISPR/Cas9 gene editing technology was used to regulate FAD2 gene expression. Firstly, the CRISPR/Cas9 single knockout vectors GmFAD2-1B and GmFAD2-2C and double knockout vectors GmFAD2-2A-3 were constructed. Then, the three vectors were transferred into the recipient soybean variety Jinong 38 by Agrobacterium-mediated cotyledon node transformation, and the mutant plants were obtained. Functional analysis… More >

Correlation Analysis of New Soybean [Glycine max (L.) Merr] Gene Gm15G117700 with Oleic Acid

    Shuo Qu1, Yaolei Jiao2, Lamboro Abraham2, Piwu Wang1,*

    Phyton-International Journal of Experimental Botany, Vol.90, No.4, pp. 1177-1192, 2021, DOI:10.32604/phyton.2021.015206

Abstract The fatty acid dehydrogenase gene plays an important role in regulating the oleic acid content in soybean. Genome-wide association study screened out soybean oleic acid related gene Gm15G117700. A fragment size of 693bp was obtained by PCR amplification of the gene and, it was connected by seamless cloning technology to the pMD18T cloning vector. Based on the gene sequence cloned, bioinformatic analysis of gene protein was performed. The overexpression vector of Gm15G117700 and the CRISPR/Cas9 gene editing vector were constructed. The positive plants were obtained by Agrobacterium-mediated transformation of soybean cotyledon nodes and T2 plants were identified by conventional PCR,… More >

Seed characteristics and fatty acid composition of castor (Ricinus communis L.) varieties in Northeast China

    Huang FL1,2, GL Zhu2,3, YS Chen1,2, FJ Meng4, M Peng1,4, XF Chen1,2, ZB He3, ZY Zhang3, YJ Chen1,2

    Phyton-International Journal of Experimental Botany, Vol.84, No.1, pp. 26-33, 2015, DOI:10.32604/phyton.2015.84.026

Abstract Oil content and fatty acid composition were investigated on 12 castor varieties and strains by using the soxhlet extraction method and capillary gas chromatography. This was made to provide a reference and theoretical basis for castorbean breeding with high oil content, determine variability of seed compounds for breeding purposes, and broaden chemical material choices. Results revealed that crude fat percentage in seeds ranged from 18.91 to 35.84% with an average of 25.91%; the absolute content of ricinoleic acid varied between 171.65 g/kg and 314.03 g/kg with an average of 222.43 g/kg, and kernel crude fat percentage was between 24.28 and… More >

Biobased Vanillic Acid and Ricinoleic Acid: Building Blocks for Fully Renewable Copolyesters

Claudio Gioia*, Maria Barbara Banella, Grazia Totaro, Micaela Vannini, Paola Marchese, Martino Colonna, Laura Sisti, Annamaria Celli

    Journal of Renewable Materials, Vol.6, No.1, pp. 126-135, 2018, DOI:10.7569/JRM.2017.634191

Abstract New fully biobased polyether/esters have been synthesized by a one-pot polymerization reaction of ricinoleic acid (RA), vanillic acid (VA) and ethylene carbonate (EC). In particular, EC selectively reacts with the phenolic group of VA to obtain in-situ 4-(2-hydroxyethoxy)-3-methoxybenzoate (EV), suitable for subsequent copolymerization with RA. The procedure was carried out in a single step, without any solvent. Chemical structure and thermal properties of the new materials were studied in order to explore relationships between composition and final performances. The combination of EV, bearing a rigid aromatic structure, with RA, characterized by high flexibility and potential biocidal activity, allows the production… More >
