YUKA IKEDA, NOZOMI NAGASE, AI TSUJI, KURUMI TANIGUCHI, YASUKO KITAGISHI, SATORU MATSUDA*

    BIOCELL, Vol.46, No.5, pp. 1163-1167, 2022, DOI:10.32604/biocell.2022.018177

Abstract Stemness of cancer cells contains limitless self-renewal proliferation. For the purpose of proliferation, secretome might exert its effects via the paracrine signaling. Specific microRNAs enclosed in the secretome of cancer stem cells could regulate the expression of anti-proliferative APRO family proteins. The biological functions of APRO family proteins seems to be quite intricate, however, which might be a key modulator of microRNAs, then could regulate the proliferation of cancer cells. In addition to affecting proliferation/differentiation during cellular development, APRO family proteins might also play an imperious role on keeping homeostasis in healthy stem cells under a physiological condition. Therefore, relationship… More >

Effect of RBM10 on regulating the proliferation and metastasis activity of human hepatocellular carcinoma cells by affecting the stability of miR-21

    LI JIANG1,*, KE ZHANG1, CHONG HU1, HONGWEI ZHANG1, DANPU WANG1, WEIWEI ZHAI2

    BIOCELL, Vol.46, No.4, pp. 969-978, 2022, DOI:10.32604/biocell.2022.017765

Abstract In recent decades, RNA binding motif (RBM) proteins have been widespread concerned by researchers. Among them, RBM5 is considered as a potential tumor suppressor gene in HCC. RBM10, also belonging to the RBM family, have similar structure and high homology with RBM5, indicating its potential as potential tumor suppressor genes. However, the role of RBM10 in tumors is controversial. The purpose of this study was to analyze the expression correlation and functional relationship of miR-21 and RBM10 in human hepatocellular carcinoma (HCC) tissues and corresponding tumor cells. Bioinformatics analysis showed that miR-21 and RBM10 were both highly expressed in HCC;… More >

    BIOCELL, Vol.46, No.3, pp. 699-709, 2022, DOI:10.32604/biocell.2022.015726

Abstract Circular RNAs (circRNAs) are a novel class of non-coding RNA that have recently shown to have huge capabilities in the regulation of gene expression at the posttranscriptional level. Growing evidence has indicated that circRNAs could serve as competing endogenous RNAs (ceRNAs) to sponge microRNAs (miRNAs) and suppress functions of targeted miRNAs. Osteosarcoma (OS) is the most common malignant primary bone cancer. Hsa_circ_0002137 is upregulated in OS. However, the role of hsa_circ_0002137 in OS remains unclear. Using miRNA pull-down assay, we showed that cir_0002137 sponged hsa-miR-1246, and BCL2 apoptosis regulator (BCL2) mRNA was a potential target of hsa-miR-1246 in human osteosarcoma… More >

STMN1 promotes the proliferation and inhibits the apoptosis of acute myeloid leukemiacells by activating the PI3K/Akt pathway

    PENG YANG1,*, ZHIYING ZOU1, XULING GAO2

    BIOCELL, Vol.46, No.1, pp. 207-218, 2022, DOI:10.32604/biocell.2021.014728

Abstract Recent studies have shown that the microtubule disrupting protein Stathmin 1 (STMN1) is differentially expressed in AML patients and healthy control. The aim of this study was to explore the effects and molecular mechanism of STMN1 in AML. Here, the expression of STMN1 in peripheral blood cells (PBMCs) and bone marrow of AML patients and healthy volunteers was detected by RT-PCR and Western blot. STMN1 expression was regulated by transfected with STMN1 overexpressed plasmid or shRNA in two human leukemia cell lines K562 and HL60. Cell proliferation was examined by CCK8 and Edu staining. Annexin V and TUNEL assays were… More >

    BIOCELL, Vol.46, No.1, pp. 171-183, 2022, DOI:10.32604/biocell.2022.015409

Abstract In order to explore the role of forkhead box protein O1 (FoxO1) in the lipid metabolism and cell proliferation, goose primary hepatocytes were isolated and incubated with insulin or PI3K-Akt-mTOR pathway dual inhibitor NVP-BEZ235, and then transfected with FoxO1 interference plasmid. The related parameters of lipid metabolism and cell proliferation were measured. The results firstly showed that FoxO1 interference increased the intracellular TG and lipids concentration (P < 0.05); and increased the proliferative index (PI), cell DNA synthesis, protein expression of Cyclin D1 in goose primary hepatocytes (P < 0.05). Secondly, the co-treatment of insulin and FoxO1 interference increased the… More >

    Çağrı Öner1,* and Ertuğrul Çolak2

    Oncologie, Vol.23, No.3, pp. 393-407, 2021, DOI:10.32604/Oncologie.2021.016958

Abstract piRNAs are novel members of small non-coding RNAs and have an impact on genetic and epigenetic mechanisms of cells. It was aimed to investigate the role of piR-651 on MCF-7 benign breast cancer cells by focusing on molecular characteristics. Anti-piR-651 was transfected and effects of piR-651 on proliferation, adhesion, and motility of MCF-7 cells were detected after the 24th, 48th, and 72nd hour. Gene expressions of piR-651, Ki-67, MMP-2, ERα, HIF-1α, and hTERT were determined by using RT-PCR. piR-651 inhibition caused the decrease of proliferation, adhesion (p < 0.001), and motility of MCF-7 cells. The efficiency of anti-piR-651 transfection supported… More >

    BIOCELL, Vol.45, No.6, pp. 1521-1526, 2021, DOI:10.32604/biocell.2021.09220

Abstract Increasing evidence proves that circular RNAs (circRNAs) play an important role in regulating the biological behaviors of tumors. The central purpose of this research was to investigate the functions of circRNA in gastric cancer. The utilization of real-time PCR was to test circPTN expression in gastric cancer cells. Cell counting colony formation assays, CCK-8 assay, and EdU assay were used to investigate proliferation. Transwell assay was applied to investigate migration. We discovered that circPTN was highly expressed in gastric cancer cells. Low expression of circPTN inhibits gastric cancer cell proliferation and migration. Elevated expression of circPTN promotes gastric cancer cell… More >

    Phyton-International Journal of Experimental Botany, Vol.91, No.1, pp. 223-238, 2022, DOI:10.32604/phyton.2022.015523

Abstract Korean pine is an important afforestation tree species in Northeast China, which has a high ecological and economic value. Although regeneration of somatic embryogenesis using immature zygotic embryos of Korean pine as explants has been successful, it cannot be applied to automation and large-scale production. Therefore, we urgently need a method that can increase the output of somatic embryos (SEs) to meet the needs of large-scale production. We used Korean pine 1–1 and 1–100 cell lines as research materials to evaluate the effects of inoculum-density, culture time, orbiting speed, vessel volume, plant growth regulator (PGR) concentration, and carbon source on… More >
