Abstract After depletion of intracellular Ca2+ stores the capacitative response triggers an extracellular Ca2+ influx through store-operated channels (SOCs) which refills these stores. Our objective was to explore if human umbilical artery smooth muscle presented this response and if it was involved in the mechanism of serotonin- and histamine-induced contractions. Intracellular Ca2+ depletion by a Ca2+-free extracellular solution followed by Ca2+ readdition produced a contraction in artery rings which was inhibited by the blocker of Orai and TRPC channels 2-aminoethoxydiphenyl borate (2-APB), suggesting a capacitative response. In presence of 2-APB the magnitude of a second paired contraction by serotonin or histamine… More >

    BIOCELL, Vol.34, No.3, pp. 121-132, 2010, DOI:10.32604/biocell.2010.34.121

Abstract Wilms tumor gene 1 (wt-1), a key regulator of mesenchymal-epithelial transformation, is downregulated during congenital obstructive nephropathy, leading to apoptosis. There is a functional interaction between WT-1 and inducible nitric oxide synthase (iNOS). In this regard, we reported that after neonatal unilateral ureteral obstruction, rosuvastatin prevents apoptosis through an increase in nitric oxide bioavailability, which in turn is linked to higher Hsp70 expression. Hence, the goal of this study was to determine whether a nitric oxide/Hsp70 interaction is involved in changes in WT-1 mRNA expression after ureteral obstruction. Neonatal rats submitted to experimental ureteral obstruction were treated with either vehicle… More >

    BIOCELL, Vol.28, No.3, pp. 287-297, 2004, DOI:10.32604/biocell.2004.28.287

Abstract As a step towards the identification of genes preferentially expressed in the oviduct during early rat embryo development, we isolated a cDNA fragment (Pr14) by using RNA arbitrarily primed PCR (RAP-PCR), being its expression restricted to oviduct and uterus; its mRNA is mainly expressed in oviduct during late luteal phase and early pregnancy. This fragment is 100% identical to a rat DNA sequence (Accession No. NW_047400) downstream the terminal exon of a Rattus norvegicus gene (Locus Link Accession No. LOC289316) similar to ebaf (endometrial bleeding-associated factor), a novel member of the Transforming Growth Factor superfamily. Northern analyses showed that this… More >

Prolactin-releasing activity of GHRP-5 (Momany peptide) on lactotrophs in vivo and in vitro

ANA LUCÍA DE PAUL*, MÓNICA BONATERRA*, CLAUDIA GABRIELA PELLIZAS**, AGUSTÍN AOKI*, ALICIA INÉS TORRES*

    BIOCELL, Vol.26, No.1, pp. 49-60, 2002, DOI:10.32604/biocell.2002.26.049

Abstract In the present study the in vivo and in vitro effects of GHRP-5 on the PRL-releasing activity in correlation with the morphological changes of lactotroph cells and their transcriptional activity were evaluated. The in vitro treatment (12 µg/100g BW/day for 3 days) of male rats with GHRP-5 does not induce any significant changes in serum PRL levels. In contrast, the addition of GHRP-5 to pituitary cell cultures increased significantly the release of PRL. This effect is enhanced in cell cultures of enriched lactotrophs, increasing significantly the secretion of PRL, the concentrations of which were 50% higher than that of untreated… More >

An integrated analysis of mRNA-miRNA transcriptome data revealed hub regulatory networks in three genitourinary cancers

    BIOCELL, Vol.41, No.1, pp. 19-26, 2017, DOI:10.32604/biocell.2017.41.019

Abstract Bladder, kidney, prostate and testicular carcinoma are the top four genitourinary cancers in China. Here we analyzed mRNA and miRNA expression profiles of carcinomas of the bladder (TCC), kidney (ccRCC) and testis (TGCT) to uncover their specific regulatory mechanisms. The gene expression profiles of GSE31617 were downloaded from GEO database, which contained 27 samples, including 10 TCC, 7 TGCT and 10 ccRCC. Specific up- and downregulated differentially expressed genes (DEGs) and differentially expressed microRNAs (DEmiRNAs) of each cancer were selected and target genes of DEmiRNAs were predicted. Gene interaction network of the shared genes and target genes of DEmiRNAs of… More >

    mRNA-specific translational regulation in yeast

    Bengü ERGÜDEN

    BIOCELL, Vol.43, No.3, pp. 103-118, 2019, DOI:10.32604/biocell.2019.06517

Abstract The expression of a gene is governed at various levels, from transcriptional to translational level. The translational control is widely used to regulate gene expression, especially when a rapid, local, and selective control over protein synthesis is required. The present review describes instructive examples of translational regulation in yeast, together with regulatory elements within mRNAs. The review also outlines the important contributions of mRNAbinding proteins that act in harmony with several translational elements to generate appropriate translational signals and responses. Mechanical Stretch-Induced Changes in Cell Morphology and mRNA Expression of Tendon/Ligament-Associated Genes in Rat Bone-Marrow Mesenchymal Stem Cells

Guanbin Song∗,†,‡, Qing Luo*, Baiyao Xu*, Yang Ju

    Molecular & Cellular Biomechanics, Vol.7, No.3, pp. 165-174, 2010, DOI:10.3970/mcb.2010.007.165

Abstract It has been demonstrated that mechanical stimulation plays a vital role in regulating the proliferation and differentiation of stem cells. However, little is known about the effects of mechanical stress on tendon/ligament development from mesenchymal stem cells (MSCs). Here, using a custom-made cell-stretching device, we studied the effects of mechanical stretching on the cell morphology and mRNA expression of several key genes modulating tendon/ligament genesis. We demonstrate that bone-marrow-derived rat MSCs (rMSCs), when subjected to cyclic uniaxial stretching, express obvious detectable mRNAs for tenascin C and scleraxis, a unique maker of tendon/ligament formation, and significantly increased levels of type I… More >
