    Oncology Research, Vol.27, No.7, pp. 779-788, 2019, DOI:10.3727/096504018X15442985680348

Abstract The aim of this study was to investigate the underlying mechanisms that transforming growth factor- (TGF- )-mediated epithelial-to-mesenchymal transition (EMT) in tumor cells contributes to 5-FU resistance. A series of experiments involving cell viability and caspase activity analyses, siRNA transfection, RNA isolation, and quantitative-PCR (qPCR) assay, cell migration analysis, Western blotting analysis of total protein and membrane protein were performed in this study. Mouse xenograft model was used to determine the effect of the PAR2 inhibitor in vivo. In this study, we found that protease-activated receptor 2 (PAR2) induction in 5-FU therapy is correlated with TGF- -mediated EMT and apoptosis… More >

    Oncology Research, Vol.27, No.3, pp. 379-387, 2019, DOI:10.3727/096504018X15199531937158

Abstract Breast cancer is one of the major malignancies with a mounting mortality rate in the world. Long noncoding RNA (lncRNA) cancer susceptibility candidate 2 (CASC2) has been identified to regulate the initiation and progression of multiple tumorous diseases according to previous studies. However, its biological role has been rarely reported in breast cancer. In the present study, lncRNA CASC2 was found to be significantly downregulated in breast cancer tissues and cell lines using real-time quantitative PCR. Furthermore, gain-offunction assays demonstrated that overexpression of lncRNA CASC2 significantly repressed breast cancer cell proliferation and metastasis. Moreover, CASC2 induced cell cycle arrest and… More >

Jun Shan Ruan*†, Huan Zhou*, Lin Yang, Ling Wang*, Zong Sheng Jiang§, Hong Sun*, Shao Ming Wang*†

    Oncology Research, Vol.27, No.5, pp. 593-600, 2019, DOI:10.3727/096504017X15051723858706

Abstract Transforming growth factor- 1 (TGF- 1)-induced epithelial–mesenchymal transition (EMT) of non-small cell lung cancer (NSCLC) may contribute to tumor metastasis. TGF- 1-induced EMT in H1975 cells (a human NSCLC cell line) resulted in the adoption of mesenchymal responses that were predominantly mediated via the TGF- 1–integrin signaling pathway. Ursolic acid has been previously reported to inhibit tumor growth and metastasis in several cancers. However, whether ursolic acid can attenuate TGF- 1-induced EMT in H1975 cells and its underlying mechanisms remain unknown. In this study, ursolic acid significantly attenuated the TGF- 1-induced decrease in E-cadherin level and elevated the level of… More >

miRNA-101 Targets TGF-bR1 to Retard the Progression of Oral Squamous Cell Carcinoma

Yong Wang*, Rui-Zhi Jia*, Shu Diao*, Jun He, Li Jia

    Oncology Research, Vol.28, No.2, pp. 203-212, 2020, DOI:10.3727/096504019X15761480623959

Abstract Despite the considerable knowledge on the involvement of microRNA-101 (miR-101) in the evolution of oral squamous cell carcinoma (OSCC), the underlying mechanisms remain obscure. In this study, miR-101 expression was markedly downregulated in the OSCC cell lines and tissues. Cell counting kit-8 (CCK-8), ethynyl deoxyuridine (EdU), and colony formation assays showed that miR-101 inhibited the proliferation of OSCC cells. Flow cytometry and caspase 3 activity assays indicated that miR-101 induced OSCC cell apoptosis. Transwell assays demonstrated that this miRNA also repressed OSCC cell migration and invasion. Moreover, tube formation assay showed that miR-101 abated the proangiogenesis of OSCC cells. Dual-luciferase… More >

Abstract Anaplastic thyroid carcinoma (ATC) is resistant to standard therapies and has no effective treatment. Eukaryotic translation initiation factor 5A2 (EIF5A2) has shown to be upregulated in many malignant tumors and proposed to be a critical gene involved in tumor metastasis. In this study, we aimed to investigate the expression status of EIF5A2 in human ATC tissues and to study the role and mechanisms of EIF5A2 in ATC tumorigenesis in vitro and in vivo. Expression of EIF5A2 protein was analyzed in paraffin-embedded human ATC tissues and adjacent nontumorous tissues (ANCT) (n = 24) by immunochemistry. Expressions of EIF5A2 mRNA and protein… More >

Abstract Thermally grown oxide (TGO) is a critical factor for the service life of thermal barrier coatings (TBC). Numerical simulations of the growth process of TGO have become an effective means of comprehensively understanding the progressive damage of the TBC system. At present, technologies of numerical simulation to TGO growth include two categories: coupled chemical-mechanical methods and mechanical equivalent methods. The former is based on the diffusion analysis of oxidizing elements, which can describe the influence of bond coat (BC) consumption and phase transformation in the growth process of TGO on the mechanical behavior of each layer of TBC, and has… More >

Knockdown of lncRNA XIST prevents the epithelial-mesenchymal transition of TGF-β2-induced human lens epithelial cells via miR-124/Slug axis

    XUE JIANG1,2, HONG ZHANG1,2,*

    BIOCELL, Vol.46, No.3, pp. 785-794, 2022, DOI:10.32604/biocell.2022.016167

Abstract Posterior capsular opacification (PCO) is linked to the pathological process of lens epithelial cells, which includes proliferation, migration, and epithelial-mesenchymal transition (EMT). Our goal was to investigate whether long noncoding RNA (lncRNA) XIST contributes to EMT via targeting miR-124/Slug axis in TGF-β2-induced SRA01/04 cells. EMT was induced by stimulating SRA01/04 cells with 10 ng/mL TGF-β2 for 24 h, and PCO microenvironment was simulated. The expressions levels of lncRNA XIST, miR-124, and Slug were measured by real-time polymerase chain reaction (RT-PCR) and western blot. The role and mechanism of lncRNA XIST in promoting EMT of TGF-β2-treated SRA01/04 cells were investigated by… More >
