Tolerance and Efficacy of Regorafenib according to UGT Pharmacogenetical Status in the Treatment of Metastatic Refractory Colorectal Cancer

    Pierre-Guillaume Poureau1,2,*, Estelle Dhamelincourt2, Jessica Nguyen2, Hélène Babey2, Emmanuelle Renaud2, Margaux Geier2, Michèle Boisdron-Celle3, Jean-Philippe Metges2

    Oncologie, Vol.23, No.2, pp. 195-202, 2021, DOI:10.32604/Oncologie.2021.015929

Abstract Introduction: Regorafenib is a multi tyrosin-kinase inhibitor prescribed in metastatic refractory colorectal cancer treatment. Its toxicity is significant but inconstant. The metabolism of regorafenib includes oxydation via cytochrome P3A4, then glucuroconjugation. A pharmacogenetical approach of mutational status of Uridine-Diphospho-Glucuronosyltransfersase (UDP-glucuronosyl-transferase, UGT) could be a strategy to optimise the use of regorafenib. Patients and Method: This is a restrospective, unicentric study. All adult patients treated with regorafenib for a metastatic colorectal cancer in our center between 2013 and 2017 were analysed. UGT1A1 polypmorphism was previously researched in the laboratory after written informed consent. Results: Thirty-five patients received regorafenib during the study… More >

Abstract Traditional Chinese medicine (TCM) has been increasingly applied in both preventing and treating a variety of cancers in the last decades, attributing to its fewer side effects as compared with chemotherapy drugs. Hellebrigenin, a component of Chanpi from the skin of Bufo bufogargarizans Cantor or Duttaphrynus melanostictus has been reported to have an obvious anti-cancer activity on various cancers. However, the effect and mechanism of hellebrigenin on colorectal cancers were still unknown. Herein, the present study demonstrated hellebrigenin significantly reduced viability and triggered apoptosis via the intrinsic pathway in colorectal cancer cell lines HCT116 and HT29 in vitro and in… More >

    BIOCELL, Vol.45, No.3, pp. 483-487, 2021, DOI:10.32604/biocell.2021.014704

Abstract The extent and aggression of colorectal cancer is a worldwide public health threat. Extensive research has been conducted on the pre-requisites leading to this fatal cancer. An array of genes along with their mutations and the signal transduction pathways leading to the cellular transformation into the cancerous cells have been investigated. Based on the knowledge gained so far, present review shortly discussed the role of the major genes especially those are involved in instigating abnormalities in the cellular cycles, cellular proliferation and differentiation. A simple but novel molecular scheme of the colorectal cancer development has also been plotted. More >

Effects of Ulinastatin Combined with Dexmedetomidine on Postoperative Cognitive Function and Central Nerve Specific Protein Level in Elderly Colorectal Cancer Patients after Laparoscopic

    Li Bao1, Xiaowei Tian2, Jing Zhang3, Li Chen4, Kui Gao5,*

    Oncologie, Vol.22, No.3, pp. 167-178, 2020, DOI:10.32604/oncologie.2020.012495

Abstract Ulinastatin combined with dexmedetomidine can improve postoperative cognitive function and central nerve specific protein (S-100β) level in elderly colorectal cancer (CC) patients after laparoscopic. Altogether 178 elderly patients who underwent laparoscopic colorectal cancer surgery in our hospital from February 2018 to August 2019 were selected and divided into two groups according to the treatment methods. Those anesthetized by dexmedetomidine were regarded as the routine group (RG, 83 cases), and those anesthetized by ulinastatin and dexmedetomidine were considered as the combined group (CG, 95 cases). The operation conditions of the two groups, the pain scores (VAS) at 4 h, 24 h… More >

Two Stage Classification with CNN for Colorectal Cancer Detection

    Pallabi Sharma1,*, Kangkana Bora2, Kunio Kasugai3, Bunil Kumar Balabantaray1

    Oncologie, Vol.22, No.3, pp. 129-145, 2020, DOI:10.32604/oncologie.2020.013870

Abstract In this paper, we address a current problem in medical image processing, the detection of colorectal cancer from colonoscopy videos. According to worldwide cancer statistics, colorectal cancer is one of the most common cancers. The process of screening and the removal of pre-cancerous cells from the large intestine is a crucial task to date. The traditional manual process is dependent on the expertise of the medical practitioner. In this paper, a two-stage classification is proposed to detect colorectal cancer. In the first stage, frames of colonoscopy video are extracted and are rated as significant if it contains a polyp, and… More >

Abstract The incidence of colorectal cancer (CRC) in China has increased in recent years. The mortality rate of CRC has become one of the highest among all cancers; CRC increasingly affects the health and quality of people’s lives. However, due to the insufficiency of medical resources in China, the workload on medical doctors has further increased. In the past few decades, the adult CRC mortality and morbidity rate dropped sharply, mainly because of CRC screening and removal of adenomatous polyps. However, due to the differences in polyp itself and the skills of endoscopists, the detection rate of polyps varies greatly. In… More >

Interaction of IL-22/IL-22R1 promotes cell proliferation and suppresses apoptosis of colorectal cancer via phosphorylation of STAT3

    Xiaoning Qin1, Liqing Yuan2, Hongxun Ruan1, Lin Lin1

    BIOCELL, Vol.43, No.2, pp. 89-98, 2019, DOI:10.32604/biocell.2019.06352

Abstract Interleukin-22 (IL-22) is a member of IL-10 cytokine family which is expressed in activated T cells predominantly and in activated natural killer cells at lower levels. Previous studies have demonstrated the link between elevated levels of IL-22 and disease severity of psoriasis, Crohn’s disease, rheumatoid arthritis and interstitial lung diseases. However, the function of IL-22 in the development and progression of colorectal cancer (CRC) remains elusive. In this study, we first evaluated the IL-22/IL-22R1 level in CRC patients, and found that tumor tissues have more active expression of IL-22 and IL-22R1 than normal tissues, presenting correlation with the degree of… More >

The expressional level of tankyrase-1 gene and its regulation in colorectal cancer in a Saudi population

    Hala Abdulaziz M ALWARTHAN1, Mohammad Saud AL ANAZI1, Narasimha R. PARINE1, Ramesa Shafi BHAT1,*, Ghadah ALAMRO1, Ftoon ALJARBOU1, Sooad K AL-DAIHAN1

    BIOCELL, Vol.43, No.2, pp. 51-58, 2019, DOI:10.32604/biocell.2019.07019

Abstract Tankyrase1 plays an essential role in cancer progression by regulating telomere length. The study aimed to determine expression of TNKS1 and its regulation in colorectal cancer (CRC) in 20 samples from Saudi patients. mRNA expression of TNKS1 in CRC and paired normal tissues was measured by qRT-PCR. Epigenetic modification of TNKS1 promoter was determined by methylation-specific PCR while somatic mutation was analyzed by Sanger sequencing in exon 10 of the gene. All cancerous and normal tissues expressed TNKS1, but level of expression in CRC tissues was significantly associated with tumor stage though no other parameters; age, gender, and tumor location,… More >
