    Technical Note : Prolonged exposure of human embryonic stem cells to heat shock induces necrotic cell death

    Abstract We investigated the effects of prolonged heat shock treatment on human embryonic stem cell (hESC) viability. The hESC viability steadily declined with longer exposure to heat shock treatment (43ºC). After 4 h of exposure to heat shock at 43ºC, only 56.2 ± 1.5% of cells were viable. Viability subsequently declined to 37.0 ± 3.3% and 3.5 ± 0.7% after 8 h and 16 h, respectively of heat shock treatment at 43ºC. Transmission electron micrographs showed that the morphology of the dead/dying cells after heat shock treatment was characteristic of cellular necrosis with an uncondensed chromatin and a non-intact plasma membrane.… More >

    Abstract Carcinoembryonic antigen (CEA) is a surface glycoprotein expressed in human epithelial cells and is released from their surface, especially during colorectal cancer. Frequently, colorectal cancer is accompanied by inflammation, where tumor-infiltrating neutrophils play an important role. CEA was also found to be a strong chemotactic agent for neutrophils. The purpose of this study was to find out if CEA can enhance neutrophil priming and activation. Primed neutrophils were activated by N-formyl-methionyl-leucyl-phenylalanine (formyl-MLP) and the resulting oxidative burst was measured luminometrically.     Abstract The goal of this study was to determine the effect of putrescine, added to the culture medium, on the in vitro development of stenospermic grape (Vitis vinifera L) embryos. The cross breedings of Perlón x G.C88552 and Perlón x Argentina were used. 0 (control), 2 and 4 mM of putrescine were added to the immature seed’s culture medium. In Perlón x Argentina, 2mM of putrescine statistically increased the percentage of total embryos, direct germination, polyembryos and normal plants.     Abstract Information on desiccation sensitivity of Euterpe edulis seeds under two drying rates is presented. The sensitivity was studied during the course of germination and normal germination. The water content was evaluated for both seeds and embryos. Results showed the following: (a) For both drying treatments and for both germination and normal germination, desiccation sensitivity values were higher for measurements based on the water content of the embryo than for those of the seed.     Abstract In this study, we established an in vitro regeneration system from immature embryos of four varieties of Sweet Sorghum with biotechnological potential.     Abstract Exposure of either gametes or embryos to conditions and/or factors that generate oxidative stress has been associated with impaired early embryogenesis. The effects of reactive oxygen species (ROS) on mouse preimplantation development, depending of the ROS-concentration and time of exposition, were studied. Two-cell embryos were incubated with 5, 10, 25 and 50 μM of hydrogen peroxide (H2O2) for 30 and 60 minutes of exposition and allowed to develop for 72 h to study the quality of development.     Abstract The aim of the current research was to clone and to characterize the partial 66 kDa streptavidin-binding peptide (SBP) found in the germinated embryos of Pisum sativum L. var. Alaska. The pea (P. sativum var. Alaska) embryos possess prominent 66 kDa SBPs that gradually disappeared after few hours of germination in germinated embryos, but not in the cotyledons.
