    Oncology Research, Vol.32, No.8, pp. 1335-1346, 2024, DOI:10.32604/or.2024.044174 - 17 July 2024

Abstract Long non-coding RNAs (lncRNAs) have been implicated in cancer progression and drug resistance development. Moreover, there is evidence that lncRNA HOX transcript antisense intergenic RNA (HOTAIR) is involved in colorectal cancer (CRC) progression. The present study aimed to examine the functional role of lncRNA HOTAIR in conferring radiotherapy resistance in CRC cells, as well as the underlying mechanism. The relative expression levels of HOTAIR were examined in 70 pairs of CRC tumor and para-cancerous tissues, as well as in radiosensitive and radioresistant samples. The correlations between HOTAIR expression levels and clinical features of patients with… More >

Abstract At present, the role of many long non-coding RNAs (lncRNAs) as tumor suppressors in the formation and development of cervical cancer (CC) has been studied. However, lncRNA prostate cancer gene expression marker 1 (PCGEM1), whose high expression not only aggravates ovarian cancer but also can induce tumorigenesis and endometrial cancer progression, has not been studied in CC. The objective of this study was to investigate the expression and the underlying role of PCGEM1 in CC. The relative expression of PCGEM1 in CC cells was detected by real-time PCR. After the suppression of PCGEM1 expression by… More > Graphic Abstract

LU ZHANG*, JINGUO CHU*, YUSHAN YU

    Oncology Research, Vol.32, No.4, pp. 703-716, 2024, DOI:10.32604/or.2023.030988 - 20 March 2024

Abstract Background: Limited research has been conducted on the influence of autophagy-associated long non-coding RNAs (ARLncRNAs) on the prognosis of hepatocellular carcinoma (HCC). Methods: We analyzed 371 HCC samples from TCGA, identifying expression networks of ARLncRNAs using autophagy-related genes. Screening for prognostically relevant ARLncRNAs involved univariate Cox regression, Lasso regression, and multivariate Cox regression. A Nomogram was further employed to assess the reliability of Riskscore, calculated from the signatures of screened ARLncRNAs, in predicting outcomes. Additionally, we compared drug sensitivities in patient groups with differing risk levels and investigated potential biological pathways through enrichment analysis, using… More >

    BIOCELL, Vol.48, No.3, pp. 387-401, 2024, DOI:10.32604/biocell.2024.048791 - 15 March 2024

Abstract Glioblastoma multiforme (GBM), the most common and aggressive primary brain tumor in adults, is the most malignant and still has no cure. However, the novel role of long non-coding RNAs (lncRNAs) in the pathogenesis of glioblastoma is attracting extensive attention. LncRNAs are transcribed RNA molecules over 200 nucleotides long that do not encode proteins. Unlike small non-coding RNAs, such as microRNAs (miRNAs), lncRNAs have more complex secondary and tertiary structures that enable them to interact with DNA, RNA, and proteins and perform multiple regulatory functions. LncRNAs act as molecular sponges, absorbing and sequestering other biomolecules,… More > Graphic Abstract

Abstract Background: Renal fibrosis is an important process in the development of chronic kidney disease. Understanding the pathogenesis and finding effective treatments for renal fibrosis is crucial. This study aims to investigate whether a newly discovered long non-coding RNA (lncRNA) called LOC103694972 could be a potential target for treating fibrosis of NRK-49F cells. Methods:: LncRNA Chip was used to identify differentially expressed lncRNAs between TGF-β1-induced NRK-49F cells and normal cells. The dual-luciferase assay confirmed the binding between miR-29c-3p and signal transducer and activator of transcription (STAT3), as well as between miR-29c-3p and lncRNA LOC103694972. Si-LOC103694972 and… More > Graphic Abstract

Abstract The role of lncRNA KCNQ1 opposite strand/antisense transcript 1 (KCNQ1OT1) in colon cancer involves various tumorigenic processes and has been studied widely. However, the mechanism by which it promotes colon cancer remains unclear. Retroviral vector pSEB61 was retrofitted in established HCT116-siKCN and SW480-siKCN cells to silence KCNQ1OT1. Cellular proliferation was measured using CCK8 assay, and flow cytometry (FCM) detected cell cycle changes. RNA sequencing (RNA-Seq) analysis showed differentially expressed genes (DEGs). Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were carried out to analyze enriched functions and signaling pathways.… More >

Abstract Nicotinamide adenine dinucleotide (NAD+) plays an essential role in cellular metabolism, mitochondrial homeostasis, inflammation, and senescence. However, the role of NAD+-regulated genes, including coding and long non-coding genes in cancer development is poorly understood. We constructed a prediction model based on the expression level of NAD+ metabolism-related genes (NMRGs). Furthermore, we validated the expression of NMRGs in gastric cancer (GC) tissues and cell lines; additionally, β-nicotinamide mononucleotide (NMN), a precursor of NAD+, was used to treat the GC cell lines to analyze its effects on the expression level of NMRGs lncRNAs and cellular proliferation, cell… More > Graphic Abstract

The relationship between lncRNA DPP10-AS1 expression in glioblastoma and patient prognosis was investigated. Cell Counting Kit-8 (CCK-8), transwell, and clonogenic experiments were utilized to assess tumor cells’ proliferation, invasiveness, and migratory potentials after lncRNA DPP10-AS1 expression was up or down-regulated. Using an online bioinformatics prediction tool, the intracellular localization of lncRNA… More >
