    Oncology Research, Vol.28, No.7-8, pp. 815-818, 2020, DOI:10.3727/096504021X16240202939988

Abstract The role of miRNAs in the radiosensitivity of glioma cells and the underlying mechanism is still far from clear. In the present study, we detected six downregulated and seven upregulated miRNAs in the serum after radiotherapy compared with paired serum samples before radiotherapy via miRNA panel PCR. Among these, miR-17-5p was highly reduced (fold change = −4.21). Further, we validated the levels of miR-17-5p in all serum samples with qRT-PCR. In addition, statistical analysis suggested that a reduced miR-17-5P level was positively associated with advanced clinical stage of glioma, incidence of relapse, and tumor differentiation. Moreover, we provided evidence that… More >

Allantoin Alleviates Seed Germination Thermoinhibition in Arabidopsis

Songbei Ying, Sasa Jing, Leheng Cheng, Haiqing Sun, Yuan Tian, Lulu Zhi, Ping Li*

    Phyton-International Journal of Experimental Botany, Vol.91, No.9, pp. 1893-1904, 2022, DOI:10.32604/phyton.2022.022679

Abstract Allantoin as the metabolite of purine catabolism can store and remobilize nitrogen for plant growth and development. However, emerging evidence suggests it also contributes to plant tolerance to stress response through altering abscisic acid (ABA) and reducing reactive oxygen species (ROS) level. 1-CYS PEROXIREDOXIN (PER1) is a seed-specific antioxidant that enhances seed longevity through scavenging ROS over-accumulation. High temperature (HT) suppresses seed germination and induces seed secondary dormancy, called as seed germination thermoinhibition. However, the mechanism that allantoin and PER1 regulate seed germination thermoinhibition remains unknown. In this study, we reported that allantoin treatment enhances seed germination under HT stress.… More >

    BIOCELL, Vol.46, No.7, pp. 1763-1770, 2022, DOI:10.32604/biocell.2022.018934

    Abstract

Drought stress is one of the major environmental obstacles that limit the production and development of apples (Malus domestica Borkh.). The role of melatonin is well known in the protection of plants under environmental stresses. In this study, we investigated the effect of melatonin on apple rootstock M. hupehensis Rehd under drought stress. The results showed that drought inhibited the growth of M. hupehensis and dramatically reduced root surface area, root volume, the number of tips and forks, and root diameter. Drought-induced growth inhibition was significantly decreased by adding melatonin. Net photosynthetic rate (Pn), intercellular CO2 concentration (Ci), stomatal conductance… More >

    HONG FU1, HAOYU ZHAO1, YALI YANG1, SIYU WANG1, KE DUAN2,*, TAILIN GUO1,*

    BIOCELL, Vol.46, No.4, pp. 1067-1078, 2022, DOI:10.32604/biocell.2022.017435

Abstract CDK5 belongs to the cyclin-dependent kinase family. CDK5 is multifunctional and plays an important role in neural differentiation. However, the role of CDK5 in osteoblastic differentiation remains unclear. The present study investigated functions and molecular mechanism of CDK5 in osteoblastic differentiation. It was found that, CDK5 inhibition promoted the expression of Runx2, ALP, OCN and OPN of MSCs and the mineralization of MC-3T3E1 cells and MSCs. CDK5 inhibition enhanced the development of F-actin, nuclear localization of β-catenin and YAP, as well as the expression of RMRP RNA. When F-actin was suppressed by Blebbistatin, the nuclear localization of YAP and β-catenin,… More >

    BIOCELL, Vol.46, No.1, pp. 137-148, 2022, DOI:10.32604/biocell.2022.016807

Abstract In this study, we investigated the protective effect of hyperbaric oxygen (HBO) on PC12 and H9C2 cell damage caused by oxygen-glucose deprivation/reperfusion and its possible mechanism. PC12 and H9C2 cell oxygen-glucose deprivation/reperfusion model were established. Cells were divided into a control group, model group, hyperbaric air (HBA) group and HBO group. The cell viability was detected by the CCK8 assay. Hoechst 33342 and PI staining assays and mitochondrial membrane potential (MMP) assays were used to detect cell apoptosis. The ultrastructure of cells, including autophagosomes, lysosomes, and apoptosis, were examined using a transmission electron microscope. The expression of autophagy-related proteins was… More >

PIWI Interacting RNA-651 Inhibition Transforms the Genetic Features of MCF-7 Breast Cancer Cells

    Çağrı Öner1,* and Ertuğrul Çolak2

    Oncologie, Vol.23, No.3, pp. 393-407, 2021, DOI:10.32604/Oncologie.2021.016958

Abstract piRNAs are novel members of small non-coding RNAs and have an impact on genetic and epigenetic mechanisms of cells. It was aimed to investigate the role of piR-651 on MCF-7 benign breast cancer cells by focusing on molecular characteristics. Anti-piR-651 was transfected and effects of piR-651 on proliferation, adhesion, and motility of MCF-7 cells were detected after the 24th, 48th, and 72nd hour. Gene expressions of piR-651, Ki-67, MMP-2, ERα, HIF-1α, and hTERT were determined by using RT-PCR. piR-651 inhibition caused the decrease of proliferation, adhesion (p < 0.001), and motility of MCF-7 cells. The efficiency of anti-piR-651 transfection supported… More >

Abstract Chemotherapy may cause cellular oxidative stress to bone marrow. Oxidative damage of bone marrow hematopoietic microenvironment is closely related to chronic myelosuppression after chemotherapeutic treatment. Angelica sinensis polysaccharides (ASP) are major effective ingredients of traditional Chinese medicine Angelica with multi-target anti-oxidative stress features. In the current study, we investigated the protective roles and mechanisms of ASP on chemotherapy-induced bone marrow stromal cell (BMSC) damage. The human bone marrow stromal cell line HS-5 cells were divided into control group, 5-FU group, 5-FU + ASP group, and 5-FU + LiCl group to investigate the mechanism of ASP to alleviate 5-FU-induced BMSC proliferation… More >

Abstract The extraction of bioactive compounds has become one of the most interesting areas of modern chemistry. For therapeutic reasons, it´s important to obtain antimicrobial agents from natural origin. The objective of the present study was to determine the inhibitory effect of ethanolic extract of basil (Ocimum basilicum L. var. Red Rubin) subjected to different concentrations of potassium (K+) on the activity of three bacterial strains that are pathogens in humans. Susceptibility was evaluated by inhibition surface and these results were compared to two antibiotics: Gentamicin (GE) and Ciprofloxacin (CPF) for their efficacy against each bacterial strain. Analyzed variables presented significant… More >

Abstract This study investigates if the anti-tumor effect of Pterostilbene in the SKOV3 ovarian cancer (OC) cell line involves inhibition of cell metabolism and tested in this effect involves modulating AMPK and Akt-induced regulation of mTORC1. Initially, SKOV3 cells were cultured in the humidified conditions in DMEM media for 24 h with or without increasing concentration of Pterostilbene. Then, the cells were incubated with Pterostilbene (IC50 = 50 µM) under similar conditions with or without pre-incubation with Dorsomorphin, an AMPK inhibitor. In a dose-dependent manner, Pterostilbene inhibited SKOV3 cell survival and increased their lysate levels of lactate dehydrogenase (LDH) and single-stranded… More >
