    BIOCELL, Vol.46, No.10, pp. 2281-2289, 2022, DOI:10.32604/biocell.2022.020697

Abstract Periodontitis is a frequent chronic inflammatory disorder destroying periodontium. Recent studies have revealed the role of bromodomain and extraterminal domain inhibitor (BETi) and microRNA (miR)-130a in regulating macrophage polarization and pro-inflammatory response. However, little is known about whether apabetalone (a novel BETi) and miR-130a are correlated with chronic inflammatory state in periodontitis by regulating macrophage polarization. Here murine RAW264.7 macrophages were applied as an in vitro inflammatory model. After treatment with Porphyromonas gingivalis-derived lipopolysaccharide (Pg LPS) and apabetalone, the expression of macrophage M1 polarization markers and inflammatory cytokines was assessed using real-time PCR, western blot, and enzyme-linked immuno sorbent assay… More >

    Oncologie, Vol.24, No.1, pp. 147-161, 2022, DOI:10.32604/oncologie.2022.019236

Abstract Tumor-associated macrophages (TAMs) are important components in tumor microenvironment. This study intended to explore the influence of TAMs on cervical cancer cells proliferation and migration. The expression levels of TAMs markers, CD68 and CD163, in tissues were examined by immunohistochemistry and increased with the progression of cervical lesions (p < 0.05). TAMs with M2-like phenotype (PMA(Polymethacrylate) induced THP-1 cells) were noticed to promote the proliferation of cervical cancer cells and improve the migration ability of tumor cells. These enhancements were attributed to secreting soluble components and the physical contact between macrophages and tumor cells. The tumor formation and tumor growth… More >

    BIOCELL, Vol.46, No.6, pp. 1505-1519, 2022, DOI:10.32604/biocell.2022.016564

Abstract The development of polycystic ovary syndrome (PCOS) is closely related to the chronic inflammatory and obese. Recent studies have found macrophages regulate the chronic inflammation and adipose tissue remodelling, but the underlying mechanisms have not been clarified. In this study, we established a model of PCOS in the offspring rats by high androgen exposure during late pregnancy in parental and established a female rat macrophage eliminating model by rejection of clodronate liposome. Then, the offspring rat macrophage phenotype in offspring female rat adipose tissue, and levels of testosterone, angiogenic factors (PDGF and VEGF) and inflammatory factors (TNF-α and MCP-1) were… More >

Human β-defensin 2 enhances IL-1β production and pyroptosis through P2X7-mediated NLRP3 expression in macrophages

    PANPAN WANG1,2, GANG LI3, LI GAO1,2, CHUANJIANG ZHAO1,2

    BIOCELL, Vol.46, No.5, pp. 1197-1207, 2022, DOI:10.32604/biocell.2022.016607

Abstract Periodontal disease is the leading cause of tooth loss, which is also a high-risk factor for other diseases including oral cancer and cardiovascular disease. Periodontitis is one of the most common type of periodontal diseases. Interleukin-1β (IL-1β) plays a key role in the pathogenesis of periodontitis. However, the mechanism how IL-1β is produced during periodontitis is still unclear. In the present study, we found that human β-defensin 2 (hBD2) enhances IL-1β production through an LPS-primed human acute monocytic leukemia (THP-1) macrophage model. Inhibition of P2X purinoceptor 7 (P2X7) reduced hBD2-enhanced IL-1β production. Incubation of LPS-primed THP-1 macrophages with potassium chloride… More >

miRNA-148b-3p targeting SOCS3 inhibits macrophage M2 polarization by JAK2/STAT3 pathway in immune thrombocytopenia

YANG YANG, LIJUAN FU, CHUNMEI CHEN, MEIWEI HU

    BIOCELL, Vol.46, No.5, pp. 1319-1328, 2022, DOI:10.32604/biocell.2022.015760

Abstract Aberrant expression of miRNAs is significantly correlated with the occurrence of immune thrombocytopenic purpura (ITP). The immune imbalance of M1/M2 macrophage contributes to the development of ITP. However, the role of miR-148b-3p in macrophage phenotype imbalance remains unknown in ITP. In this study, we aimed to explore whether miR-148b-3p inhibits M2 macrophage polarization in ITP and to investigate the underlying mechanism. Peripheral blood from 22 ITP patients were collected, and real-time PCR confirmed that miR-148b-3p was up-regulated and Western blot analyses detected the expression of SOCS3 was down-regulated. Subsequent dual-luciferase reporter gene assay indicated that miR-148b-3p could bind to SOCS3.… More >

Abstract Differentiated macrophages have been proven to participate in the development of mesenchymal stem cells in different tissues. However, the regulatory processes remain obscure. Exosomes, which are key secretions of macrophages, have attracted increasing attention. Therefore, macrophage-derived exosomes may modulate the development of Bone marrow mesenchymal stem cells (BMMSCs). Different culture conditions were used to induce M1 polarization in THP1 cells. Subsequently, exosomes derived from unpolarized (M0) and polarized (M1) macrophages were isolated, BMMSCs were cultured with normal complete medium or inductive medium supplemented with M0 or M1 derived exosomes, and the osteogenic capacity of the BMMSCs was measured and analyzed.… More >

    BIOCELL, Vol.44, No.4, pp. 623-629, 2020, DOI:10.32604/biocell.2020.012645

Abstract Macrophages play an essential role in the myocardial ischemia-reperfusion injury (MIRI), and the macrophage shifting from M1 to M2 phenotypes might be a potential strategy for the treatment of MIRI. It has been reported that miR-182 plays an important role in MSC-Exo-associated macrophage polarization. As circBCRC-3 is a newly discovered circle RNA that worked as a sponge of miR-182, this research aimed to find if circBCRC-3 plays a role in MSC-Exo-associated macrophage polarization. Firstly, circBCRC-3 was identified by divergent primers in mesenchymal stem cells (MSCs). Secondly, the exosome of MSCs was isolated and identified by transmission electron microscopy (TEM), nanoparticle-tracking… More >

Abstract The rapid emergence of multidrug-resistant and extensively drug-resistant Tuberculosis retrieved intense interest in phage-based therapy. This old approach, which was abandoned in the west in the 1940s but is generating renewed interest, has stimulated fresh research on mycobacteriophages and their lytic efficiency against their hosts. GP30 is a novel protein of the mycobacteriophage CASbig with undiscovered function. In this study, we analyzed the role of CASbig gp30 in the host Mycobacterium smegmatis. Overexpression of gp30 in the host led to reduced growth in acidic medium and attenuated the intracellular survival rate of M. smegmatis inside the THP-1 macrophages, which may… More >
