Summary
HER2-directed treatment has changed considerably in recent years. Initially associated mainly with breast cancer, HER2 is now investigated and used as a therapeutic target in a wider range of solid tumors. Its amplification, overexpression, or mutation has been reported in gastric, colorectal, lung, biliary tract, and gynecological malignancies, although the clinical significance may differ from one tumor type to another.
This Special Issue will discuss the current and emerging role of HER2-targeted therapies beyond breast cancer. We are particularly interested in studies focused on monoclonal antibodies, antibody-drug conjugates, tyrosine kinase inhibitors, and treatment combinations. Another important area is the interpretation of HER2 testing, especially in tumors where the optimal diagnostic approach and therapeutic thresholds are still under discussion.
Original studies, reviews, and clinically oriented perspectives are welcome. Topics may include patient selection, treatment sequencing, mechanisms of resistance, tumor heterogeneity, safety, and real-world outcomes. The aim is to bring together evidence from different malignancies and to clarify how HER2-directed strategies can be integrated into personalized cancer care.
