Summary
Human papillomavirus (HPV) infection is a major cause of infection-related malignancies. It is associated with a broad spectrum of mucosal lesions and cancers affecting different anatomical sites, including the oral and oropharyngeal, anal, cervical, vulvar, vaginal, and penile regions. Persistent infection with high-risk HPV types can promote malignant transformation through the activity of viral oncoproteins and their interactions with cellular signaling pathways, host immune responses, and the tumor microenvironment. However, HPV persistence and progression from infection to precancerous lesions and cancer are influenced by anatomical, viral, and host-related factors. This Special Issue aims to provide a multidisciplinary overview of HPV-associated malignancies, focusing on the mechanisms underlying viral persistence, HPV-driven carcinogenesis, lesion progression, and malignant transformation across different anatomical sites. Particular attention will be given to host–virus interactions, immune dysregulation and immune evasion, molecular pathways, biomarkers of infection and disease progression, and factors influencing treatment response and resistance. Contributions addressing HPV-associated lesions in different anatomical sites, immunocompromised populations, immunotherapy, targeted therapies, therapeutic HPV vaccines, and emerging strategies for cancer prevention, early detection, and treatment are particularly encouraged. By integrating clinical, molecular, and translational perspectives, this Special Issue seeks to advance the understanding of HPV-driven carcinogenesis and identify new opportunities for risk assessment, prevention, and therapeutic management of HPV-associated malignancies.
