Summary
Multidrug resistance (MDR) remains one of the most formidable obstacles in successful cancer treatment, frequently leading to therapy failure and disease relapse. A major contributor to MDR is the overexpression of ATP-binding cassette (ABC) transporters, a family of membrane proteins capable of actively effluxing a wide range of chemotherapeutic agents out of cancer cells. Despite decades of research, the clinical targeting of ABC transporters has yielded limited success, and their roles extend far beyond drug efflux, encompassing functions in cell signaling, metabolism, immune response, and the tumor microenvironment. This Special Issue aims to bring together original research and review articles addressing the multifaceted roles of ABC transporters in cancer biology and therapy resistance. Topics of interest include but are not limited to: molecular mechanisms of ABC transporter regulation; interactions with oncogenic pathways; novel modulators or inhibitors; experimental models to study transporter activity; epigenetic, transcriptomic, or post-translational regulation; implications for immunotherapy or targeted therapy; and clinical relevance as prognostic or predictive biomarkers. We welcome contributions that explore innovative experimental approaches, translational insights, or therapeutic strategies aimed at overcoming ABC transporter-mediated resistance in cancer. The ultimate goal is to provide a comprehensive and updated overview of the field, opening new avenues for improving cancer treatment outcomes.
