Summary

This special issue proposal discusses pivotal aspects of diagnosis, multidisciplinary care, and therapy developments with a focus on head and neck cancer. The goal of the issue is to give a thorough review of the most cutting-edge methods and best practices being used in the industry today.

Modern diagnostic methods will be examined, with an emphasis on how they affect early diagnosis and better patient outcomes. We also aim to explore how important it is for multidisciplinary teams to work together to optimize treatment plans and enhance patient care. Case studies and expert perspectives will be used to analyze this collaborative approach, presenting effective examples of interdisciplinary teamwork. Lastly, articles on the most recent therapeutic developments, such as innovative treatment modalities and their efficacy in managing different stages of head and neck cancer, will be included.

The ultimate objective is to offer a current and pertinent selection of clinical insights and research that will be helpful to patients, researchers, and physicians alike, greatly advancing the ongoing management of head and neck cancer.

Abstract Head and neck squamous cell carcinoma (HNSCC) is an aggressive cancer with high recurrence rates and prevalent resistance to therapeutic interventions. Tumor behavior is largely dependent on the tumor microenvironment (TME) that includes immune cells, stromal components, cancer-associated fibroblasts (CAFs), the extracellular matrix (ECM), and an associated cytokine network. In this review, we examine principal mechanisms of the tumorigenic transformation, encompassing immune checkpoint disruption, therapy resistance mediated through CAFs, the contribution of hypoxic niches, and several metabolic dependencies that hold potential as future targets. Novel therapeutics developed and/or repurposed, such as immune checkpoint inhibitors (ICIs),… More >
