Summary

The development of treatment resistance and treatment-related toxicity remains a major challenge in cancer therapy, limiting therapeutic efficacy and affecting patients' quality of life. Although promising advances in immunotherapy, targeted therapies, precision medicine, antibody–drug conjugates, and novel drug-delivery approaches have transformed cancer treatment and improved outcomes for many patients, the emergence of therapeutic resistance and off-target toxicity remain major challenges in clinical practice and patient care. This Special Issue aims to highlight emerging strategies for overcoming therapeutic resistance and reducing or preventing adverse effects associated with anticancer treatments. We welcome original research and review articles investigating the molecular and cellular mechanisms underlying drug resistance and treatment-induced toxicities, as well as novel approaches to improve treatment efficacy and safety. Topics may include chemotherapy resistance, targeted therapy and immunotherapy resistance, chemotherapy-induced toxicities, predictive biomarkers, drug combinations, drug repurposing, natural compounds, novel drug-delivery systems, and personalized therapeutic approaches. The Special Issue will provide a multidisciplinary platform for innovative strategies that can enhance the therapeutic potential of anticancer treatments while minimizing toxicity.
